Effect Of Zheng’s Zhu Gui-san On Hemorheology And Histomorphology In SD Rats Of Acute Soft Tissue Injury

Objective: To observe the variation of the Symptomatology index,to test the Hemodynamic index, and to analysis the Muscular Histomorphology is a project to explore the impact of Zheng’s Zhu Gui-san in acute soft-tissue injury. Therefore, the aims of this experimental study are to assess the effect of Zheng’s Zhu Gui-san on promoting the injury healing. Then use the results to provide reliable experimental and theoretical basis for clinical treatment of acute soft-tissue injury.Method:72 SD rats with acute soft-tissue injury were divided into 3 groups: Placebo group(with Saline, n=24), Qili-san group( produced by Tong Ren tang pharmacy,n=24), and Zheng’s Zhu Gui-san group. Acute soft-tissue injury was induced on the right hind limb gastronemius using a percussive device. Drugs stared to use after muscle contusion was induced. Every 6 rats from one group were killed on the 1th 、3th、7th and 15 th day after using different drugs. The Hemorheology index including whole blood viscosity(WBV)、Whole Blood reduced viscosity(WBRV)、plasma viscosity(PV) 、 HCT 、 erythrocyte aggregation index(EAI) 、 erythrocyte deformability index(EDI) and FIB would be analysis by testing the sample of blood from femoral artery of SD-rats. As well as the Damage Symptomatology index(DSI) and Muscular Histomorphology(MH).Results: Damage Symptomatology index(DSI): The first day after modeling, There was no significantly different among these three groups(P>0.05); On the third day of after modeling, compared with the placebo group, the DSI of Zheng’s Zheng’s Zhu Gui-san group and Qili-san group were significantly decrease after contusion(P<0.05). However, there was no significant different between Zheng’s Zhu Gui-san group and Qili-san group(P>0.05). On the 7th day and 10 th day after modeling, the DSI was marked decline in bothe Zheng’s Zhu Gui-san group and Qili-san group. But, the index was gently descend in placebo group. Both Zheng’s Zhu Gui-san group and Qili-san group had significant different Comparing with placebo group(P < 0.05). Otherwise, there was no significant different between Zheng’s Zhu Gui-san group and Qili-san group(P > 0.05). Hemodynamic index: Day1, among whole blood viscosity(WBV)、Whole Blood reduced viscosity(WBRV)、plasma viscosity(PV)、HCT、erythrocyte aggregation index(EAI) and FIB in Zheng’s Zhu Gui-san group and Qili-san group had seen different level of increasing without remarkable differences. Besides, the erythrocyte deformability index(EDI) had decreased in the same time(P>0.05). Day3, the WBRV、the PV、the HCT 、the EAI and the FIB has marked sink in group Zheng’s Zhu Gui-san and Qili-san, there was significant different compare with the group of placebo(P<0.05); But, there is no statistic significant between Zheng’s Zhu Gui-san group and Qili-san group(P>0.05). All of the Hemorheology index in the Zheng’s Zhu Gui-san group and Qili-san group had been substantially decreased in the day 7th and 10 th after modeling, except EDI When it comes to compare to placebo group. The EDI index had been increased as well(P<0.05).Muscular Histomorphology: No significant differences in Muscular Histomorphology had appeared among three kind of groups in the first day after modeling(P>0.05). From the third day to the tenth day after modeling, group Zheng’s Zhu Gui-san and Qili-san had remarkable differences when comparing with placebo group(P<0.05). However, there was no significant difference between Zheng’s Zhu Gui-san group and Qili-san group in every time point. Conclusion(1)Zheng’s Zhu Gui-san can rapidly relieve the symptom after acute soft-tissue injury like redness、swelling、heat and pain;(2) The injury can be effectively treated by improving the viscosity of blood with using Zheng’s Zhu Gui-san; the healing function related to reduce plasma viscosity, improve Erythrocyte rheology, restrain FIB appearing and the function of antiplatelet aggregation;(3) Zheng’s Zhu Gui-san has a clear therapeutic effect on rats with acute soft-tissue injury.
