Objective: This study establish an acute injury model of soft tissue of SD rats by weight injury,and use Jindao-San to therapy the injury by gavage.Observed experimental rats histomorphology,soft tissue injury symptomology,local skin temperature regularly,detection the content of PGE2 in local tissue homogenate To explore the Jindao-San's effects on acute soft tissue injury in SD rats and the possible mechanism of treatment,and provide a theoretical basis for the clinical treatment of acute soft tissue injury of the drug.Methods: A total of 96 three months old healthy male SD rats were selected and randomly grouping sections were divided into four groups(n = 24): the experimental group(Jindao-San group),the drug control group(Qili-San),the model control group and blank control group.The first three groups were modeled by heavy weight injury.The blank control group only shaved off the hair of the leg without modeling.Then each group was randomly divided into four time points(1 day group,3 day group,7 day group,10 days group,n = 6).After modeling,the model were given experimental drugs and equal volume of saline gavage.At the each time point recording weight of the rats;measuring bilateral leg skin temperature;scoring soft tissue damage symptom index,and building tissue slices of the modeling marked area,then observe the histomorphology under light microscope and using enzyme-linked immunosorbent assay detected the concentration of prostaglandin E2 in tissues;Using the statistical software(SPSS 19.0)to analysis data.Results:(1)Tissue slice observation,rating:On the 1st day,Jindao-San group,drug control group,model control group had significant difference compared with the blank control group(P < 0.01),there is statistical significance.On the 1st day and the 3rd day,Jindao-San group,drug control group compared with model control group had statistically difference(P < 0.05),but there is no obvious difference between Jindao-San and drug control group(P > 0.05).On the 7th days and the 10 th day,Jindao-San group,drug control group compared with model control group had statistically difference(P < 0.05),but there is no obvious difference between two groups too(P > 0.05).On the 10 th day drug control group,the Jindao-San group and model control group compared with the blank control group P < 0.05,the difference to be markedly,with statistical significance.(2)Symptomatology score: There was no statistically difference(P> 0.05)between the Jindao-San group,the drug control group and the model control group at the 1st and 3rd day after modeling.On the 7th day,the model control group are statistically difference between the Jindao-San group and drug control group(P < 0.05);On the 10 th day,Jindao-San group and the drug control group compared with model control group,the P value was less than 0.01,the difference was highly statistically significant.The Jindao-San group and the drug control group at each phase comparison,no statistically difference(P > 0.05).(3)local skin temperature difference value:On the 1st day after modeling,Jindao-San group,drug control group,model control group rats skin temperature difference value between the statistics has no obvious difference(P > 0.05).On the 3rd and 7th day,the Jindao-San group,drug control group compared with model control group had statistically difference(P < 0.05);But no obvious difference between two groups(P > 0.05).On 10 th day,Jindao-San group,drug control group,model control group rats skin temperature almost returned back to normal,compared between three groups,the difference is not obvious,no statistical significance(P > 0.05).(4)PGE2 content: On 1st day after the modeling,model control group PGE2 content in the tissue homogenate compared with blank control group PGE2 content are highly significant difference(P < 0.01);In comparison with the model control group PGE2 content found that On 1st,3rd,7th,10 th day Jindao-San group and drug control group PGE2 content with statistically difference(P < 0.05),Jindao-San group PGE2 content compared with drug control group,was no statistically difference between the two groups(P > 0.05).Conclusion:(1)The rat model of soft acute tissue injury was successfully made by striking the injury(2)Jindao-San treatment of soft tissue acute injury SD rats have a clear effect.The effect of Jindao-San on PGE2 content in acute tissue injury may be reduced by reducing the level of local tissue damage to PGE2,with anti-inflammatory,analgesic and antipyretic effects.
