| Objective: Zheng's Xiaozhongzhitong-Tang,which from the traditional Chinese medicine orthopedic specialist Zheng Huaixian.In the Zheng's theory clearly pointed out that the prescription has blood,swelling,analgesic effect for the soft tissue,especially for pain of swelling.In this study,we observed and recorded the changes of acute soft tissue injury syndrome,histology,local tissue and hypothalamic?-endorphin in SD rats.And compare with common prescription Qili-San and Ibuprofen sustained release capsules.To evaluate the curative effect of Zheng's Xiaozhongzhitong-Tang on soft tissue acute injury and to explore its therapeutic mechanism,and to provide animal test support for the clinical application of Zheng's Xiaozhongzhitong-Tang.Methods:Healthy three-month-old SD rats 96,weighting 363.40±4.0g.After 7 days of adaptive feeding,the rats were shaved off the bilateral buttocks and bilateral thighs on the day before modeling.The markers were marked with 1 cm on the bilateral thighs to determine the position of the model and the position of the bilateral skin temperature.Select the left side of the rat marked mold,and select the successful model of 80 rats,random marking after weighing.Group A: model control group,Group B: drug control group 1(ibuprofen group),Group C: drug control group 2(Qili-San group),Group D: experimental group(Zheng's Xiaozhongzhitong-Tang group).After the modeling and grouping,the drug dose required for the experiment was calculated according to the method of human and animal drug dose conversion.The experiment group was filled with decoction and the drug control group was filled with turbid suspension.Model control group was filled with serving saline.Free to drink water.To observe the hematopoietic status of the left lower limb model of the rats,the degree of swelling of the skin and the activity disorder.At the same time,the specimens were taken at 8:00 on the 1st,3rd and 7th day,and the specimens were observed under light microscope.And homogenate of hypothalamus,and the concentration of ?-endorphin was detected by enzyme-linked immunosorbent assay.The results are processed by software for data analysis.Results:(1)Body weight Body weight of four groups of rats slightly changed.Model group,Ibuprofen group and Zheng's Xiaozhongzhitong-Tang group showedan increasing trend,in group of Zheng's Xiaozhongzhitong-Tang the first time point compare with the fourth time point P<0.05.There are no differences between the two groups in statistics.(2)Body temperature Four time points in the four groups of rats in the morning temperature and nocturnal temperature lower extremity local temperature gradually decreased.There were no statistically significant differences in the morning and nocturnal temperature difference between the four groups(model group,Ibuprofen group,Qili-San group,Zheng's Xiaozhongzhitong-Tang group).(3)Symptom index Four groups of syndrome index were gradually decreased.In the second and third point,the difference between the model group and other three groups was statistically significant(P <0.01);in the fourth point,the difference between the model group and Qili-San group,Zheng's Xiaozhongzhitong-Tang group was statistically significant(P <0.01),and the difference between the Ibuprofen group and Qili-San group,Zheng's Xiaozhongzhitong-Tang group was statistically(P<0.05).(4)?-endorphin Model group hypothalamic ?-endorphin content in the slow rise at the second time point reached the peak then decreased.Ibuprofen group gradually decreased after reaching its peak at the first point.Qili-San group gradually increase from the first time point to the fourth time point to reach the maximum.Zheng's Xiaozhongzhitong-Tang group gradually increased at the second time point to reach the peak followed by a slight decrease.In the first point,the difference between the model group and other three groups was statistically(P <0.05);In the second point,the difference between the Zheng's Xiaozhongzhitong-Tang group and other three groups was statistically(P <0.05),In the fourth point,the difference between the model group and Qili-San group was statistically(P <0.05).Four groups of rat tissue ? – endorphin.Model group slowly increased at the third time point to peak and then decreased,Ibuprofen group gradually decreased after reaching its peak at the first point,Qili-San group gradually decrease from the second time point to the peak,Zheng's Xiaozhongzhitong-Tang group gradually increased in the second time point to reach the peak after a slight fluctuation.In the first and second point,the difference between the model group and other three groups was statistically(P <0.05),In the fourth point,the difference between the model group and Qili-San group was statistically significant(P <0.01).(5)Histomorphological score In the second point,the difference between the model group and other three groups was statistically significant(P <0.01),In the third and fourth point,the difference between the model group and other three groups was statistically(P <0.05)Conclusion:1 Zheng's Xiaozhongzhitong-Tang and Ibuprofen,Qili-San,as are the acute soft tissue injury have a better effect,can effectively reduce the local tissue swelling,slow down the pain of local tissue.2 Zheng's Xiaozhongzhitong-Tang in the third day to improve the hypothalamic endorphins,local muscle tissue endorphins,and thus participate in the body's pain relief. |
