Objective:In the aim of exploring the therapeutic effect on acute soft tissue injury and possible mechanism of Zheng's Danggui Powder?ZDP?,further improving Zheng's injury prescription system,laying a solid theoretical foundation for its subsequent research and development and clinical application,the effects of ZDP on the changes of syndromes,hemorheology,and histomorphology in rat acute soft tissue injury were observed and recorded.Method:Eighty-four SPF 7-week-old male Sprague-Dawley rats were selected and used to establish an acute soft tissue injury model by beating the right hind limb of SD rats.First,they were randomly divided into three large groups after successful modeling:Zheng's Danggui Powder Group?ZDPG?,Qili Powder Group?QPG?and saline group?SG?according to the random number table method.Each large group consisted of 24 experimental rats and several spare rats.Second the rats of each large group were divided into 4 groups of 6 mice each in the same way.The experimental rats and relevant sapre rats of each group received the same treatment.Third,the three groups were given the corresponding medications every day.Forth,after the above steps,some monitoring indicators were observed,measured and scored daily:the general condition of the SD rat,the body weight,the skin temperature and the damage symtomatology index?DSI?of the soft tissue injury.Others were detected on the 1st,the 3rd,the7th and the 12th days after the modeling day:histomorphometric and hemorheological parameters.The histomorphometric parameters were detected by grading the histologic section of the local damaged muscle tissue.The hemorheological parameters were gained by detecting the blood collected from the abdominal aorta of SD rats,including whole blood viscosity?WBV?,whole blood reduced viscosity?WBRV?,hematocrit?HCT?,plasma viscosity?PV?,fibrinogen?FIB?,erythrocyte aggregation index?EAI?and erythrocyte deformation index?EDI?.Results:?1?Body weight:The body weight of SD rats in each group showed an upward trend with time.There was no significant difference in body weight when compared each group at the time of each material collection?P>0.05?.?2?Skin temperature:The skin temperature difference between the healthy and affected sides?STD?of SD rats in each group showed a decreasing trend over time.There was no significant difference in STD between the three groups on the first day?P>0.05?.On the 3rd day,QPG had no significant difference in STD compared with SG and ZDPG?P>0.05?;Compared with SG,the STD of ZDPG was significantly decreased?P<0.05?.On the 7th and 12th days,ZDPG and QPG had significantly lower STD than SG?P<0.05?,but there was no significant difference between ZDPG and QPG?P>0.05?.?3?Damage symtomatology index:The DSI in each group showed a decreasing trend over time.There was no significant difference between the three groups on the first day?P>0.05?.On the 3rd day,QPG had no significant difference in DSI compared with SG and ZDPG?P>0.05?;Compared with SG,the STD of ZDPG was significantly decreased?P<0.05?.On the 7th and 12th,the DSI of QPG and ZDPG were significantly lower than that of SG?P<0.05?,but there was no significant difference between ZDPG and QPG?P>0.05?.?4?Histomorphometric parameters:The histomorphometric parameters of the SD rats in each group showed a decreasing trend over time.There was no significant difference between the three groups on the first day?P>0.05?.On the 7th and 12th,QPG had no significant difference in histomorphometric parameters compared with SG and ZDPG?P>0.05?;Compared with SG,the histomorphometric parameters of ZDPG was significantly decreased?P<0.05?.On the 12thh day,the parameters of SG and QPG had no significant difference?P>0.05?,but the parameters of ZDPG were significantly lower than those of SG and QPG?P<0.05?.?5?Hemorheological parameters:In all groups,except the EDI showed an increasing trend over time,the other hemorheological parameters of SD rats showed decreasing trend.On the first day,there was no significant difference in all hemorheological parameters between the three groups?P>0.05?.On the third day,compared with QPG,ZDPG had significantly lower WBV?in high shear situation?and HCT?P<0.05?,and significantly higher EDI?P<0.05?,except the other hemorheological parameters;Compared with SG,ZDPG and QPG had significantly higher EDI?P<0.05?,and other parameters were significantly decreased?P<0.05?.On the 7th day,compared with QPG,ZDPG had significantly lower WBV?in low shear situation?and HCT?P<0.05?,and significantly higher EDI?P<0.05?,except the other parameters;Compared with SG,ZDPG and QPG had significantly higher EDI?P<0.05?,and other parameters were significantly decreased?P<0.05?.On the 12th day,there was no significant difference between ZDPG and QPG in all hemorheological parameters?P<0.05?,but compared with SG,ZDPG and QPG had significantly higher EDI?P<0.05?,and other parameters were significantly decreased?P<0.05?.Conclusion:This study use ZDP to cure the acute closed soft tissue injury.Based on the observation of physical signs,symptoms,histologic sections and the detection of hemorheological parameters,the conclusion can be drawed as follows:First,ZDP can significantly improve the typical symptoms of inflammation of the SD rats after acute soft tissue injury,such as redness,swelling,hot and pain.Second,ZDP has an obvious anti-inflammatory analgesic effect because of it can effectively alleviate muscle tissue edema and hyperemia and reduce inflammatory cells infiltration.Third,ZDP can significantly improve the hemorheology and change the level of blood viscosity.Besides,it has the effect of anticoagulation,inhibiting erythrocyte aggregation and platelet aggregation.All the effects of ZDP can help improve local blood circulation and promote tissue recovery and healing.
