Objective: Zheng’s Zhugui-San is clinical experience of famous Wu Yi people of Professor Zheng Huaixian, that have promoting blood circulation,detumescence and analgesic effects in his book "Chinese orthopedic treatment experience". In this experiment, observe and record oral Zheng’s Zhugui-San on SD rats with acute closed soft tissue injury recovery after injury affected, morphologic changes, and changes in homogenates of interleukin 6, and with Qili san(Beijing Tong Ren Tang Tong Ren Tang pharmaceutical Co., Ltd.) for comparison of treatment effects,treatment for comparison, discussion of Zheng’s Zhugui-San on acute closed soft tissue injury treatment, and for the application to provide theoretical basis for clinical treatment of acute soft tissue injury.Research methods: The 75 two-month-old, male rats were made model, selected the successful model of 72 rats,and randomly labeled with a marker then weighing and recording the weight.according to the random number table divided into three groups(n=24), group A: blank control group, group B:drug control group, group C : drug experimental group. Zheng’s Zhugui-San, Qili san made turbid suspension and administered orally drug experimental group and drug control group by gavage method, the blank control group fed with normal saline, 2 times a day. Then each group was randomly divided into 4 time point group(1, 3, 7, 10, n=6). At each observation time point were observed in skin swelling, bruising petechiae and activities; observed and evaluation the morphological changes under the light microscope;to detect tissue interleukin 6 content by enzyme immunoassay(ELISA);and the results using the software for data processing and analysis.Results:(1)Weight Results: The three groups, the blank control group weight is relatively stable over time gradually increased; Qili San group’s weight has dropped in first 7 days, 7 days after the rise; and Zheng’s Zhugui-San group rats at 4 time points over time, body weight continued to rise;Comparison between the three groups, the 1、3day, the three groups of rats were not significantly different; the 7 day, Zheng’s Zhugui-San group and blank control group、Qili San group were significant difference(P<0.05); the 10 day, Zheng’s Zhugui-San group and blank control group with a difference(P<0.05), Zheng’s Zhugui-San group and Qili san groups had no difference(P>0.05);(2) symptoms index of soft tissue injury:the index of three groups showed a continued downward trend. After modeling the first and third days of damage index score three groups of rats no significant difference; after modeling seventh days Zheng’s Zhugui-San group and blank control group were significantly different(P < 0.05),Zheng’s Zhugui-San group and Qili san group were not statistically significant; tenth days after modeling, blank control group and Zheng’s Zhugui-San group、Qili San group had significant difference(P < 0.05), Zheng’s Zhugui-San group and Qili San group had no significant difference;(3)Histological score: Over time, the three groups of rats histological scores have declined, Qili San group and Zheng’s Zhugui-San group than in the blank control group dropped more quickly. Comparison within groups: the blank group after modeling for seventh days, tenth days compared with third days had significant difference(P < 0.05); Qili San group 7th days after modeling there was a significant difference compared with first days, third days, 10 th days compared with the 1st day, 3rd days, there was statistical significance; Zheng’s Zhugui-San group compared 7th days with 1st days there was statistically significant, the 10 th days and the 1st day、3rd days 、7th days had significant difference. Comparison between groups: first days after modeling, the comparison between three groups had no significant difference. Third days, seventh days, tenth days of Zheng’s Zhugui-San group, Qili san group compared with the blank control group there were significant differences, the Zheng’s Zhugui-San group and Qili san group was not statistically significant at each time point.(4)The content of IL-6(ng/L): Within group comparisons showes: there was no significant difference between the blank control group at each time point. Qili San group of first days and third days, seventh days, tenth days there were significant differences(P<0.05).Zheng’s Zhugui-San group first days compared with seventh days、tenth days was statistically significant(P <0.05).the comparison between groups: first days after modeling, control group,Qili san group, Zheng’s Zhugui-San group had no significant difference; third days after modeling, Qili San group compared with the blank control group with significant difference(P<0.05), there was a significant difference Zheng’s Zhugui-San group than the control group(P<0.05), Qili San group and Zheng’s Zhugui-San group had no significant difference. After modeling for seventh days, Qili San group compared with the blank control group with significant difference(P < 0.05), Zheng’s Zhugui-San group and blank group, Qili San group showed no significant difference.Tenth day of modeling, the blank group, Qili San group, Zheng’s Zhugui-San group was no significant difference.Research conclusions: Zheng’s Zhugui-San for acute soft tissue contusion have a therapeutic effect, can reduce the amount of inflammatory cytokines IL-6 local tissue damage by inhibiting the activity of inflammatory cytokines to reduce damage to the local tissue inflammation the reaction thus shortening the treatment of acute soft tissue injury time.
