Objective: To observe the Jindao-San's effect in treating acute soft tissue injuries in mice.The effect was evaluated by its role in inflammatory processes and the recovery of soft tissues.The injury syndrome index,body temperature,morphology and IL-1? levels were detected.This study would provide theoretical evidences for the application of orthopedic prescription descended from Zheng Huaixian.Methods: A total of 96 SD rats were randomly divided into the Jindao-San group,the Qili-San group,the model control group,the blank control group.Acute soft injuries model were established in mice by blowing the soft tissues heavily.Intragastric administrations were performed 1h after successful establishment of the soft injury model.Injury syndrome index,temperature of both the injured and the uninjured limb were measured at day 1,3,7,10 after remodeling.The morphology was observed via staining of the injured tissue and IL-1?levels were detected by ELISA.The data were analyzed by SPSS22.0.Results:(1)The observation of the injury symptom index: On the 1 day after modeling,the Jindao-San group,the Qili-San group and the model control group,were in the most serious damage stage,then the injury symptom index began to debilitate,until the end of the experiment.After modeling,no statistical significance was observed at day 1,dya3,P> 0.05.Compared with the model control group,the Jindao-San group and the Qili-San group were significantly different in 7,10 days,P<0.05 and P<0.01.At each time point,there was no significant difference between the Jindao-San group and the Qili-San group,P>0.05.(2)Injured skin temperature: the Jindao-San group,the Qili-San group and the model control group started to rise from the day 1,peaking at day 3,then gradually decline until the end of the experiment.After modeling,no significant difference was observed at day 1,day 10,P> 0.05.Compared with the model control group,the Jindao-San group and the Qili-San group were significantly differentat day 3 and day7 P<0.05.At each time point,there was no significant difference between the Jindao-San group and the Qili-San group,P>0.05.(3)The observation of tissue morphology: The damage of three groups was all trend that the local inflammatory cell infiltration decreased,the tissue edema alleviate and gradually repaired with the passage of time.After modeling,no significant statistical difference was observed,P> 0.05.Compared with the model control group,the Jindao-San group and the Qili-San group were significantly different,at day 3,day 7 and day 10.P<0.05.At each time point,there was no significant difference between the Jindao-San group and the Qili-San group,P>0.05.(4)the level of IL-1?: the Jindao-San group,the Qili-San group and the model control group started to rise from day 1,peaking at day 3,then gradually decline until the end of the experiment.Compared with the blank control group,the modeled three groups were significantly different in 1 days,P<0.05;Compared with the model control group,no significant difference was observed in the Jindao-San group at day 1,P>0.05;the Qili-San group was significantly different in 1 days,P<0.05.Compared with the model control group,the Jindao-San group and the Qili-San group were significantly different at day 3,day 7 and day 10,P<0.05.At each time point,there was no significant difference between the Jindao-San group and the Qili-San group,P>0.05.Conclusion:Jindao-San can effectively improve the signs and symptoms in SD rats with acute soft tissue injury,including local fever,bruising,swelling and movement dysfunction incurred by injury.It can also reduce the level of IL-1?,inhibit the excessive inflammatory reaction and then effectively promote the rehabilitation of the injured tissue.Jindao-San and Qili-San have the similar effect on the treatment of acute soft tissue injury.
