Full Field Optical Coherence Tomography (FFOCT) is a biological imaging technique with micrometer-scale resolution. it has the capability of yielding the high resolution images of biological tissue noninvasively, overcoming the some disadvantages of the traditional OCT such as low imaging speed, high prices and the low resolution.Several key problems in FFOCT imaging were investigated in this thesis. First of all, based on the review of the imaging characteristics and the limitations of the traditional OCT, we had analysised the imaging characteristics and System architecture of FFOCT. Secondly, the imaging propertios and the system configurations of FFOCT were discussed, including the light source, the illumination system, and the interferometer. The simulation of the performance of the system was also performed. We had builded the FFOCT system and obtained the quite clear interferometric images with light emitter diode LED as the light source. Thirdly, in order to eliminate the nonlinearity, a four steps phase shift method was used to re-built the structure of the sample. In order to obtain four frames of the interferometric images with pi/2 phase difference, a CY8C29466 was used to control the PZT, A program for rebuilding the three-dimensional structure of the sample was also designed. Finally, a filter based on the CY8C29466 of PSoC with central frequency 26KHZ,bandwidth 12KHZ, was designed and tested. The work in this thesis is an important part of the FFOCT system setting up in our Lab.
