| Objective:In this study,we studied the effect of Bushen Zhuyun Decoction on ovarian manganese superoxide dismutase(SOD-2)and glutathione peroxidase in rat model of Luteal phase defect(LPD)induced by mifepristone.The effects of(GPX-1),P53 mRNA and protein levels,Bax and Bcl-2 on morphological changes of rat ovary in LPD model,and preliminary study on the mechanism of Bushen Zhuyun Recipe in treating LPD.Methods:50 female SD rats of 8 weeks old were randomly assigned to the blank group,mifepristone group,positive drug dydrogesterone group,Bushen Zhuyun prescription low dose and high dose treatment group,a total of 5 groups,each Group 10 only.Among them:the blank control group was treated with isotonic equal volume of normal saline;the mifepristone group:treated with mifepristone for 2 personality cycles;the positive drug for dydrogesterone group:treated with mifepristone for 2 individual cycles,Prednisolone tablets treatment 2 personality cycle;Bushen Zhuyun prescription low-dose and high-dose group:treated with mifepristone 2 personality cycle,to the kidney to help the pregnant side to deal with 2 personality cycle.The ovarian structure was observed by HE staining.The expression levels of SOD-2,GPX-1 and P53 protein in ovary were detected by Western blot.The ovarian SOD-2,GPX-1 and P53 were detected by polymerase chain reaction(PCR),Bcl-2,BAX mRNA expression levels.The above data were statistically analyzed using SPSS22.0 statistical software.Results:1.Morphological comparison of ovaries in each group(1)HE staining of ovary in the mifepristone group showed more sinusoid follicle vacuolation,granulosa cell reduction,and disordered arrangement in the ovarian tissue;follicular atresia at all levels,residual follicles became irregular,and ovarian interstitial tissue Structural disorder.(2)After administration of dexamethascone and Bushen Zhuyun Decoction at low dose and high dose,a certain number of primordial follicles,anterior sinus and sinus follicles were observed in the ovarian tissue structure.The pristone group is dense and regular.2.Comparison of ovarian SOD-2,GPX-1 protein and mRNA expression levels in each group(1)Compared with the control group:the expression levels of SOD-2 and GPX-1 protein and mRNA in the ovary of the mifepristone group were significantly decreased(P<0.05);(2)Compared with mifepristone group:the expression levels of SOD-2 and GPX-1 protein and mRNA in ovary of rats with low dose and high dose of Bushen Zhuyun Recipe increased in different degrees(P<0.05);There was no significant improvement in the level of SOD-2 mRNA in the creatinine group(P>0.05).3.Comparison of P53 protein and mRNA,BAX and Bcl-2 mRNA expression in ovary of each group(1)Compared with the control group:the expression of P53 protein and mRNA and the expression of apoptosis-related factor BAX mRNA in the ovary of the mifepristone group were significantly increased(P<0.05),while the expression of anti-apoptotic factor Bcl-2 mRNA was observed.The level was significantly decreased(P<0.05);(2)Compared with the mifepristone group:the expression of P53 protein and mRNA and the expression of BAX mRNA in the ovary of the rats in the low-dose group and the dydrogesterone group were significantly decreased(P<0.05);The expression of anti-apoptotic factor Bcl-2 mRNA in the low-dose group increased significantly(P<0.05).However,the expression of P53 protein and mRNA and BAX mRNA in the high-dose group of Bushen Zhuyun Recipe decreased to different degrees.The expression level of apoptotic factor Bcl-2 mRNA also increased,but there was no statistically significant(P>0.05).However,the expression level of anti-apoptotic factor Bcl-2 mRNA in dydrogesterone group did not improve significantly.Conclusions:Bushen Zhuyun Recipe can improve the structure and morphology of ovary in LPD model rats.In terms of mechanism,Bushen Zhuyun Recipe may improve the anti-oxidation and anti-apoptotic levels of ovarian granulosa cells in LPD model rats and reduce the ovarian granulosa cells.The level of death reaches the goal of treating LPD. |
