| Objective: This study is to discuss the therapeutic effect and possible mechanism about Zheng'SXiaoZhongZhiTongTang treatment of acute soft tissue injury and to provide a certain basis for clinical medication,by establishing the animal model and observing the changes of local IL-6 concent and tissue morphology recovery from our institute's late famous Sports Medicine Specialist Professor Zheng Huaixian's experience prescription Zheng'SXiaoZhongZhiTong Tang after acute soft tissue injury.Methods: Three months 80 male SD rats are selected and randomly divided into four groups(n=20)after molding,another spare group of 16 rats without any processing.With Zheng'SXiaoZhongZhiTong Tang as a treatment drug,the drug should be taken two times every day after the success molding for an hour,the ibuprofen group and the Qilisan group as the positive control group.Rats in each group are sacrificed in the four phases,1,3,7,10 days after molding.Five rats in each group in every phase are randomly selected and sacrificed,and four rats of spare group are selected and sacrificed in the first day,before executing observing and recording the Symptomatology score after injury.Then along 1 cm injury part of the rat hips back from the popliteal fossa as the center,removal of the muscle is about 1.5 cm by 1.5 cm by 0.5 cm.One half of the sample is into 10% formaldehyde solution to fix,detecting the content of IL-6 with the ELISA method.The other half with foil package and marking is in the refrigerator of-70 to save.After doing all ?this,sample should be paraffin wax slices and tissue section HE staining in time and do Histomorphology observation under light microscopy.The study will conclude the treatment effect of Zheng'SXiaoZhongZhiTongTang on acute soft tissue injury and the possible mechanism of promoting tissue repair,by analyzing four groups of rats after acute soft tissue injury four phases' semiotics symptomatology,histological change and the content of IL-6 in local tissue.Results: IL-6 is in a same level in four rats local soft tissue of the normal group.In the model control the content of IL-6 of rats local soft tissue is increasing continually in the two phases,1,3 days,but decreasing in the two phases,7,10 days after injury.In the ibuprofen group and the Qilisan group,local content in rats is the highest in the first day,decreasing continually in the 3,7,10 days after injury,and the peak value is below the highest point of the the model group.In the Zheng'SXiaoZhongZhiTongTang,the content of IL-6 is increasing continually in the 1,3 days,decreasing continually in the 7,10 days after injury,decreasing continually in the 7,10 days after injury,and the integral level of IL-6 in the third day is below the peak value of the model group,the ibuprofen group and the Qilisan.The content of local IL-6 of four groups of rats is higher than the normal group.In the Zheng'S XiaoZhongZhiTongTang,the ibuprofen group and the Qilisan group,the content of tissue homogenate IL-6 is lower than the model control group in the 1,3,7,10 days.The local tissue IL-6 content of Zheng'SXiaoZhongZhiTongTang in the first day after injury compared with the ibuprofen group,the Qilisan group and the model control group,there is a significant difference(P<0.01).Compared with the ibuprofen group and the Qilisan group after injury in the third day,there has no statistical difference(P>0.05).Compared with the ibuprofen group and the Qilisan group after injury in the 7th day,no significant difference(P>0.05).Compared with the ibuprofen group and the Qilisan group after injury in the 10 th day,there has no statistical difference(P>0.05).Zheng'SXiaoZhongZhiTongTang,ibuprofen group and Qilisan group in 1,3,7 three phases after injury compared with model control group,there is a significant difference(P<0.01).Zheng'SXiaoZhongZhiTongTang,the ibuprofen group and the Qilisan group in the 10 th day after injury compared with the model control group,there is a significant difference(P<0.05).Conclusion: Zheng'SXiaoZhongZhiTongTang not only has the obvious anti-inflammation,but also can better promote the repair effect of soft tissue injury.One of the mechanism of the curative effect of treating soft tissue injury is connected with production of inflammatory cytokines IL-6 which regulates local soft tissue. |
