Objective:In this experiment, based on the SD rats with acute closed soft tissue contusion, using our school late renowned Professor Martial arts and medical expert Zheng huaixian' clinical experience prescription as a new experimental drug. Compared with the classical drugs for the treatment of injuries Zheng Gu Shui (National Drug Approval No. Z45021659 Guangxi Yulin pharmaceutical Co., Ltd.). Through soaking the experimental drug, Observe the repair of acute injury in rats injured limb;Simultaneous detection of the NO content of damaged tissue and observed the changes of histological. To explore the effect and the treatment mechanism of Zheng's XinShangYaoShui on the treatment of acute soft tissue injury. Provide theoretical support for its better service of clinical.Methods:Buy no special pathogen (SPF) level healthy SD rats,80, Make modle of acute closed soft tissue injury by the Blunt blow way. Select 72 accord with standard of model rats, Weighing, recording and number its. Divided into three groups according to random number table method(n=24).The experimental group (group of Zheng's XinShangYaoShui), drug control group (group of Zheng Gu Shui), blank control group (normal saline group).Select four time points of 1,3,7,10day after making modle,every time point shares 6 rats (n=6). Soak the experimental group, drug control group, blank control group respectively by Zheng's XinShangYaoShui, Zheng Gu Shui and normal saline group, twice a day. Weighed at each time point, The blind method was used to evaluate the Symptoms index of soft tissue injury, Nitrate reductase method was used to determine the concentration of NO in muscle tissue. Observed of tissue morphology by 400 times light microscope and scored. The statistical software (SPSS21.0) was used to deal with the analysis of relevant data.Results:(1) Weight Results:In this study, three groups of four time points no significant difference in body weight (P> 0.05), three groups of rats and there was no significant difference in the body recombination (P> 0.05), not experimental drug impact on the rat model of diet.(2) Symptomatology Rating: ?On the first day of the model and the first day after modeling, there was no statistically significant difference between the three groups (P> 0.05);?After modeling third, seven, ten days, three groups of rats damage symptoms were gradually reduced, but the increase of the experimental group's and the drug control group's were more rapidly than the control group's.The scores of Zheng's Xin Shang Yao Shui group and the Zheng Gu Shui group had no significant difference (P> 0.05) and they were significantly different (P< 0.05) with the normal saline goup.(3) Histological observation:Compare various time points, the most serious time of edema and inflammatory response was the first day. With the passage of time,the edema and inflammatory reaction was reduced and the Inflammatory cell infiltration decreased.As similar as the Zheng Gu Shui group,the Zheng's Xin Shang Yao Shui group was more rapidly than the normal saline goup in the improvemen of tissue morphology,and the effect was more obvious. From the point of view of morphological scores:on the first day after modeling, between the three scores showed no significant difference (P> 0.05); On the 3,7,10days,there were no significant difference between the experimental group and the drug control group (P> 0.05),and they both had significantly different (P< 0.05) with the blank control group.(4) NO content detected:1) between the two groups: ? on the first day after modeling, the NO content of experimental group and the drug control group were lower than the blank control group, they were both significantly different (P<0.05) with the blank control group,and they had no significant difference (P between groups> 0.05) with each other.?On the third,seven,ten days, NO content in three groups reached the peak,the Zheng's Xin Shang Yao Shui group's and the Zheng Gu Shui group's were lower than the normal saline goup's,they both had significantly different with the blank control group(P< 0.05).2) Within group comparison: There were significantly differentce between the third day and the ten day in the blank control group (P<0.05); There were significantly differentce between the third day and the seven day in the Zheng's Xin Shang Yao Shui group(P<0.05); There were no significantly differentce between the four time points in the Zheng Gu Shui group (P> 0.05).Conclusion:Zheng's Xin Shang Yao Shui and Zheng Gu Shui both would improve the symptoms of acute soft tissue injury and inflammatory reaction. At the same time, they could also prevent the increase of the concentration of NO in the local tissue during the acute phase of inflammation, and promote the repair of tissue damage.
