Objective: Zheng's XinShang-Yaoshui is a clinical experience of late famous Professor Zheng Huaixian, The book " Zheng's Theory and clinical of Traumatology " records it has promoting blood circulation,detumescence and analgesic effects.In this experiment, observe and record Zheng's Xin Shang-Yaoshui on SD rats with acute closed soft tissue injury recovery after injury affected, morphologic changes,and detect the content of TNF-? in serum., and comparing with Zheng Gu-Shui's treatment effect. To explore the therapeutic effect of Zheng's Xin Shang-Yaoshui for acute soft tissue injury,and provide theoretical basis for the research and clinical application of Professor Zheng Huaixian's Traumatology drug system.Methods: The80 three-month-old, male rats were made model, selected the successful model of 72 rats,the rest is Spare rats,and randomly labeled with a marker then weighing and recording the weight.According to the random number table divided into three groups(n=24),group A:drug experimental group(Zheng's Xin Shang-Yaoshui),group B:drug control group(Zheng Gu-Shui), group C:blank control group(normal saline).Rats were placed in glass bottles and then received the treatment of drugs for external use. the Purpose of glass bottles were to prevent the rat agonizeover,bite and destroy topical drugs.Medication for 2 hours, two times a day.Daily observation, whether the rats have skin allergies or other adverse reactions. Meanwhile, each group was randomly divided into 4 time point group(1, 3, 7, 10, n=6). At each observation time point were observed in skin swelling, bruising petechiae and activities, observed andevaluation the morphological changes under the light microscope, detect the content of TNF-? in serum.by enzyme immunoassay(ELISA),Finally using statistical software SPSS20.0 for data processing analysisResults:(1) Weight Results: After first day and third day of modeling, the weight of the three groups were reduced, and then gradually increased to the maximum of 10 th day, Comparing between the three groups, the the weight of blank control group was the lightest in the10 th day,But the result of the weight in the three group were no significant differences(P>0.05).(2)Soft Tissue Injury Index:On the first day after modeling,Three groups were the most serious damage, And then the Symptomatology index showed a continued downward trend. After modeling,each group has no significant difference in1 st day(P> 0.05).by the test,the 3th day,7th day,10 th day, Zheng's Xin Shang-Yaoshui group, Zheng Gu-Shui scores were obviously lower than the blank control group and with a significant difference between the blank control group(P<0.05),but there was no significant difference between Zheng's Xin Shang-Yaoshui group and Zheng Gu-Shui(P>0.05).(3) Histological observation: Observation of the tissue under light microscope,as time goes on, from third day, tissue edema of Zheng's Xin Shang-Yaoshui group and Zheng Gu-Shui group of began to ease, vascular congestion were relieved,infiltration of inflammatory cells were reduced, Showing a trend of the reduction of inflammatory response,but the blank group was significantly reduced from 7th day.Microscopic data score results:Zheng's Xin Shang-Yaoshui group, Zheng Gu-Shui group were significantly better than the blank control group, Zheng's Xin Shang-Yaoshui group, Zheng Gu-Shui scores were lower than the blank control group,and with a significant difference between the blank control group(P<0.05). on the 3th day,7th day and 10 th day, whereas no significant difference between Zheng's Xin Shang-Yaoshui group and Zheng Gu-Shui(P>0.05).(4) TNF-? content(pg/ml): Within group comparisons showes: The TNF-? content of the blank control group was the highest on the first day,significantly reduced from 7th day, by the test, 7th day, 10 th day has significant differences compared with first day(P< 0.05). Zheng's Xin Shang-Yaoshui group and Zheng Gu-Shui group values from the third day decreased significantly, by the test,3th day,7th day, 10 th day tow group both has significant differences compared with first day(P< 0.05);the comparison between groups: first days after modeling, the content of three group were no significant differences(P > 0.05), form the 3th day,7th day, 10 th day, Zheng's Xin Shang-Yaoshui group, Zheng Gu-Shui scores were lower than the control group group,and with a significant difference(P<0.05),but no significant difference between Zheng's Xin Shang-Yaoshui group and Zheng Gu-Shui(P > 0.05).Contusion:Zheng's Xin Shang-Yaoshui have a therapeutic effect for acute soft tissue injury, It can reduce the signs and symptoms of acute soft tissue injury and reduce the content of serum TNF-?, Inhibit the excessive inflammatory reaction and relieve pain,then effectively promote the repair of damage.It's effectiveness is similar to Zheng Gu-Shui,And no skin irritation or other adverse reactions were found.
