Effect Of Yangyin Runmu Pill On The Survival Rate Of Lacrimal Gland Cells And The Expression Of P53 Gene In Dry Eye Rabbits

Objective:To explore the mechanism of Yangyin Runmu Pill on the growth of lacrimal gland cells in dry eye rabbits and its role in the signal transduction pathway with p53 as the core.Method:1.15 healthy New Zealand rabbits with a body weight of 1.5 ± 2.0kg were selected.The daily dosage of drug was calculated by referring to the table of ratio of common experimental animals to human body surface area.After randomly grouping,rats were respectively given distilled water,Yangyin Runmu pill and Qiju Dihuang pill,twice a day,for 7 days,fasting for 12 hours before blood collection,and taking blood from abdominal aorta after anesthesia with 20% urethane 1.5g/ml,The serum containing drugs was prepared by high speed centrifugation.2.The lacrimal gland epithelial cells were isolated by trypsin digestion after anaesthesia.The culture system was established and identified by immunohistochemistry.The growth of lacrimal gland epithelial cells was observed by inverte d microscope.3.TNF-?(2.5ng/ ml)was added into the normal culture medium to construct the inflammation microenvironment and prepare the stable rabbit stem eye cell model.4.After the second generation lacrimal gland epithelial cells were stained with fluorescein,the cell morphology was observed under fluorescence microscope,the cell survival rate was detected by MTT method,and the expression of p53 gene was detected by real time PCR method.Result:Compared with inflammatory factor intervention group A,the cell morphology of Yangyin Runmu pill group was more regular,necrotic cells decreased,Yangyin Runmu pill can effectively promote the growth of lacrimal gland epithelial cells,inhibit the expression of p53 gene.Conclusion:Yangyin Runmu pill can improve the morphological structure oflacrimal gland,promote the differentiation of lacrimal gland cells,regulate its growth,and down regulate the expression of p53 gene.It is suggested that the mechanism of Yangyin Runmu pill in treating dry eyes may be to inhibit the expression of TNF-?or p53 gene.The effectiveness of Yangyin Runmu pill in treating dry eyes was confirmed from the experimental level.
