Objective: To study the effects of Testosterone Propionate on the viability,apoptosis and Bcl-2,Bax expressions of rabbit lacrimal gland epithelial cells which were affected by H2O2,in order to investigate it's mechanism of treating dry eye syndrome in vitro .Methods: 1. The isolation of rabbit lacrimal gland epithelial cells was adopted by modified enzyme digesting.2. Various concentrations of Testosterone Propionate(1×10-8mol/l,1×10-7mol/l,1×10-6mol/l,1×10-5mol/l)were added and incubated for 24 hours, and then H2O2 was added and incubated for 60 minutes. Cell viability was detected with MTT colorimetric assay. Cell apoptosis was detected by TUNEL technology.3. Various concentrations of Testosterone Propionate(1×10-8mol/l,1×10-7mol/l,1×10-6mol/l,1×10-5mol/l)were added and incubated for 24 hours, and then H2O2 was added and incubated for 60 minutes. Bcl-2 and Bax were detected by immunohistrochimical staining.Results: 1. The acquiring rate for rabbit lacrimal epithelial gland cells were 6×106 -8×106 cells per lacrimal gland with the purity of more than 90% and the viability of more than 96%.2. Various concentrations of Testosterone Propionate(1×10-8mol/l,1×10-7mol/l,1×10-6mol/l,1×10-5mol/l)significant increased LGCs viability,as well as inhibited the apoptotic rate of lacrimal gland epithelial cells in dose dependent as compared to group B.3. Compared to group B, Bcl-2 expression was increased in lacrimal gland epithelial cells treated by various concentrations of Testosterone Propionate(1×10-8mol/l,1×10-7mol/l,1×10-6mol/l,1×10-5mol/l), Bax expression was decreased.Conclutions:1. The result shows that the isolation and culture of rabbit lacrimal gland epithehlial cells adopted by modified enzyme digesting is successful. 2. Testosterone Propionate can inhibit lacrimal gland epithelial cells apoptosis induced by H2O2.3. The mechanism of apoptosis of rabbit lacrimal gland epithelial cells inhibited by Testosterone Propionate might be modulated by increasing Bcl-2 and decreasing Bax.
