| Objective:To investigate the expression of gasdermin protein D(GSDMD),a Pyroptosis execution protein,and the serum inflammatory factor interleukin-1β,18(IL-1β,18)in a rabbit model of dry eye.And the effects of different concentrations of Yangyin Runmu Pill on the levels of gasdermin protein D(GSDMD),and i nte r le u ki n-1β、18(IL-1β and 18),serum infla m matory factors,in the conjunctiva of rabbits with dry eyes,and to find the pathways of action of Ya ngyin Runmu Pill in the treatment of dry eyes.Methods:Forty New Zealand rabbits of both sexes were selected and divided into a blank group(group A),a model group(group B),and low,medium and high dose groups of Yangyin Runmu Pill(groups C,D and E).The latter 4 groups were subjected to topical drops of 0.1%benzalkonium chloride solution in two eyes for modeling for 21 days,twice a day,and after successful modeling,the drug intervention group was subjected to different concentrations of Ya ngyin Runmu Pilll by gavage,and the material was taken for testing after 14 days.GSDMD expression in the conjunctiva was detected by immunoblotting,serum inflammatory factors IL-1β and 18 levels were detected by ELISA,and the cellular morphology of the conjunctival epithelium was observed under transmission electron microscopy.Results:1.The re were significant differences in the tear film rupture time(BUT)and(tear secretion test)SIT reduction between the model and Yangyin Runmu Pilll groups before and after modeling(P<0.05),and there was no difference in the comparison between groups(P>0.05),the cornea had different degrees of staining,and the cellula r structu re of the conj u nctival epithelium was damaged,and the model was established.2.After treatment,BUT,SIT,and FL in groups C,D,and E increased to different degrees compared with those before treatment,which was statistically significant(P<0.05).After treatment,SIT elevation in group E was significantly different com pa red with C and D,and the re was no significant difference in groups C and D(P>0.05).There was a significant difference in the improvement of BUT in group C compared with groups D and E after treatment(P<0.05),while there was no significant difference between groups D and E(P>0.05).The difference in corneal staining between groups after treatment was significant(P<0.05).3.After treatment,the ultrastructure of the conjunctival epithelium in the three groups of Yangyin Runmu Pill improved com pa red with the model group,with the most obvious improvement in group E.4.After treatment,the levels of GSDMD protein and serum inflammatory factors IL-1β and 18 in the conjunctival epithelium of the 3 groups of Ya ngyin Runmu Pill improved com pa red with the model group(P<0.05).Conclusion:A rabbit model of dry eye can be successfully established using 0.1%benzalkonium chloride topical drops for 21 days.Yangyin Runmu Pill can increase tear secretion,improve tear film rupture time and improve corneal fluorescein staining in rabbits with dry eyes.It can down-regu late the expression of GSDMD protein in the conjunctival epithelium and reduce the level of IL-1β and 18,with the best effect in the high-dose group. |
