| Objective: To investigate the effect of Yangyin Runmu Pill on the expression of p53 protein,p53 phosphorylation and p53 acetylation in conjunctival epithelial cells of dry eye rats.Methods:Fifty Sprague-Dawley(SD)male rats were randomly divided into 5 groups:blank control group(A),sham operation group(B),model group(C),Yangyin Rumu group(D),and Qijudihuang Pill(E),10 in each group.For the model group,Yangyin Runmu Pill group and Qijudihuang Pill group,castration model was manufactured by castration method.One week after the model was successfully established,each rats was given intragastric administration.Before and 1 month after drug administration,the tear secretion test(SIT)and tear breakup time(BUT)were measured in each group.Conjunctival tissue changes were observed under light microscope and the conjunctival epithelium was measured by Western-Blot for Phosphorylation of p53 protein,p53 phosphorylation,and p53 acetylation.Results: Compared with the model group,tear secretion was significantly increased and tear film rupture time was prolonged in the Yangyin Runmu Pills group.Compared with the model group,the conjunctival epithelium of the rats in the Yangyin Runmu Pill group was arranged more neatly,and the cell morphology was more regular and inflammatory,and necrotic cells decreased;Yangyin Runmu Pills can effectively reduce the expression of p53 protein,p53 acetylation,and p53 phosphorylation in the conjunctival epithelium of dry eye model rats,and the difference was statistically significant(P<0.01).Conclusion: Yangyin Runmu Pills have improved ocular surface symptoms,increased tear secretion,prolonged tear film rupture time,repair of conjunctival epithelial tissue,reduced p53 protein,p53 acetylation,p53 phosphorylation in conjunctival epithelial cells of dry eye model rats.The role of expression.It is suggested that the pathogenesis of dry eye is closely related to apoptosis regulated by p53 signaling pathway.The treatment of dry eye with Yangying Runmu pill has broad prospects. |
