Objective (1) prepared plasma containing Buddleja Officinali Flavone. (2) cultivated Lacrimal gland epithelial cells and established Cell model of dry eye in vitro. (3) Observation the Influence of drug-containing plasma on the lacrimal gland epithelial cells and expression of Bax mRNA Bcl-2 mRNA in Lacrimal gland epithelial cells.METHODS (1) 20 rats(male,healthy,0.2kg) divided into A,B,2 groups averagely and randomly;the Drug-containing plasma group (A),the blank plasma group(B),intragastric administration with Buddleja suspension and normal saline respectively,2times/d,blooding after 7 days,obtained plasma containing Buddleja Officinali Flavone and blank plasma. (2) cultivated Lacrimal gland epithelial cells and established Cell model of dry eye in vitro. cell fusion, the cultured cells were divided into three groups randomly,Buddleja Officinali Flavone group, Androgen group,blank group, Intervened with Buddleja Officinali Flavone plasma,Testosterone propionate and blank plasma. Intervention 48h, then were added to a final concentration of 100μmol/l H2O2 continued for 60 minutes, induce apoptosis. (3) TRIzol reagent extracted the total RNA. RT-PCR method to detect the expression of apoptosis-related protein of Bax mRNA and bcl-2 mRNA in the lacrimal gland epithelial cells.Results (1) Successfully gaind drug-containing plasma and blank plasma,and be utilizd to Lacrimal gland epithelial cells in vitro. (2) Successfully cultured the Lacrimal gland epithelial cells. (3) Buddleja group and androgen group can inhibit the expression of Bax mRNA in the lacrimal gland epithelial cells, compared with the'blank group significantly (P<0.01), the expression of Bax mRNA of Buddleja group Lower than the androgen group, the difference was significant (P<0.01), the expression of Bcl-2 mRNA of Buddleja group and androgen group were higher than the blank group, the difference was significant (P <0.01), the Bcl-2 mRNA of Buddleja group was higher than the androgen group, the difference was significant (P<0.01).Conclusion (1) the plasma containing Buddleja Officinali Flavone can inhibit the apoptosis of lacrimal gland epithelial cells. (2) The cell model of dry eye which have lower androgen with plasma containing Buddleja Officinali Flavone can inhibit the expression of Bax mRNA, the expression of Bax mRNA was reduced, while promoting the expression of Bcl-2 mRNA, the Bcl-2 mRNA expression increased. (3) plasma containing Buddleja Officinali Flavone inhibit apoptosis of Lacrimal gland epithelial cells possible through the channels of Inhibition the expression of Bax mRNA and promotion the expression of Bcl-2 mRNA.
