The Effect Of Yangyin Runmu Wan On The Expression Of Bcl-2 And Bax Total Protein In Lacrimal Gland Cells Of Dry Eye Rabbits

Objective:To study the effect of Yangyin Runmu Pill on the expressio n of Bcl-2 and Bax in lacrimal gland epithelial cells of dry eye rabbi ts.To analyze the effect of Yangyin Runmu Pill on the apoptosis of la crimal gland epithelial cells in dry eye rabbits,and to provide theoreti cal basis for the clinical use of Yangyin Runmu pill in dry eye treat ment.Methods:1.Preparation of serum,15 healthy New Zealand rabbits were r andomly divided into four groups:blank group,model group,treatment g roup,control group,5 in each group.After one week of adaptive feeding,the patients were treated by gavage for one week.Blood samples werecollected from abdominal aorta of 15 experimental rabbits after localdisinfection.After centrifugation,the separated serum was stored at-20? for cold storage.2.The lacrimal gland cells were isolated and ide ntified.The lacrimal gland tissues of rabbits were extracted under ase ptic condition.The lacrimal gland epithelial cells of rabbits were cultu red in vitro and the growth of the cells was observed under microsco pe.TNF-?(2.5NG/ml)was added to the normal culture medium to obs erve the model of tear gland cell injury induced by TNF-?.The expres sion of bc1-2 and Bax protein in lacrimal gland epithelial cells of ra bbits was measured by wstemnr blot.Results: 1.By isolating and culturing lacrimal gland epithelial cells,su bculture and identification,respectively,LPS and TNF-? were used to in tervene in the establishment of the model,and the biological characteri stics of the two models were compared.Finally,TNF-? inflammatory fa ctors were used to establish the stem eye cell model.2.Compared wit h each group,the drug containing plasma of Yangyin Runmu pill can promote the expression of Bcl-2 protein and inhibit the expression ofBax protein.Conclusion: Yangyin Runmu pill can effectively inhibit cell apoptosis,possibly by regulating the level of apoptosis factors Bcl-2 and Bax protein,which proves that cell apoptosis is closely related to the develop ment of dry eye.
