The rabbit,as a member of common animals for experiment,represents an important role in medical field.Safe anesthesia is required in the process of scientific research and surgical treatment in rabbits.As one of aspects,it is ensured that no effect can be found under the anesthesia for the intention of the test.Besides,a safe anesthesia with appropriate depth is required for clinical treatment.However,it still exists challenge for safe anesthesia,such as large fluctuations in cardiopulmonary parameters,and long maintenance in anesthesia.Also,the depth of anesthesia and the anesthetic dose both exist problems to solve.Inhalation anesthesia brings good effects and controllable depth,while it requires expensive cost and special equipment,injection anesthesia is more appropriate.Combined anesthesia has been the mainstream of anesthesia in rabbits.However,a safe anesthetic for rabbits is still under development.The aim of the essay is to develop a kind of safe and effective anesthetic protocol for rabbits.All animals were administered with the protocol determined in the pre-experiment:dexmedetomidine(90 ?g·kg-1)and midazolam(0.90 mg·kg-1).The protocol was applied for monitoring basic cardiopulmonary parameters(rectal temperature,respiratory rate,heart rate),biopsy activity,anesthetic effect,special monitoring(arterial blood oxygenation,noninvasive blood pressure,arterial blood gas),and laboratory monitoring(blood biochemistry,blood routine)at different time points in rabbits of anesthesia.Meanwhile,sterilization,gastrectomy,and common carotid artery bypass surgery were operated to further verify the effects of anesthetic protocol.The results showed that the experiment had a rapid induction of 2.21±0.57 min.Animals lost consciousness and muscle relaxation in the anesthetic period of 45.38±7.52 min.Besides,no excitement was appeared during the revival of 51.24±4.57 min,and the rabbits showed all a stable waking period.The anesthesia effect was good,and all the sedation,analgesia and muscle relaxation were balanced.The rectal temperature declined.Respiratory rate and heart rate dropped firstly but latterly increased within normal physiological ranges.The oxyhemoglobin saturation maintained over 93%.Blood pressure,arterial blood gas,and blood routine were all varied within physiological ranges,which indicated that it had a slight impact on the respiratory and circulatory system function.Blood biochemical indicators were in the normal physiological range,indicating that the liver and kidney function was mild.No obvious side effects had been found.Confirmatory surgeries were all operated successfully.In conclusion,the protocol of dexmedetomidine and midazolam can be applied for rabbit anesthesia which obtained a smooth and rapid induction and good sedative,analgesic,muscle relaxation effect.Less impact was found in physiological function.It can satisfy the need of relevant areas in rabbit research and clinical diagnosis and treatment in anesthesia.
