Oral anesthesia refers to the way in which narcotic drugs are absorbed and administered orally for anesthesia. It mainly acts through the absorption of gastrointestinal mucosa. In the process of oral anesthesia, using the gel matrix as the drug carrier can make the drug adhere to the oral cavity for a short time before being ingested. If the selected anesthetics have high permeability in the oral mucosa, part of the anesthetics can be rapidly absorbed through the oral mucosa to exert their efficacy, thereby shortening the induction time of anesthesia to a certain extent.Oral anesthesia has the characteristics of wide application range and high safety, which can reduce the stress response of animals during anesthesia. It is of great significance in the management of stray animals, and can also be used as a pre-medication for pet inspections, which is convenient for veterinarians to complete outpatient inspections or assist in trauma treatment. In this experiment,dexmedetomidine,zoletil,clonidine,α-chloralose were used to screen suitable anesthetic drug formulations through direct oral anesthesia, and then the appropriate anesthetic doses for direct oral and oral gel anesthesia were screened respectively, verify the practicability and safety of two oral compound anesthesia protocol, and provide experimental basis for clinical anesthesia in cats.Test content:(1)Screening of anesthetic drug prescriptions: α-chloralose,clonidine,Dexmedetomidine,and Zoletil were combined respectively, and the anesthetic prescriptions with less side effects on cats were screened out by direct oral administration.(2)Screening of anesthesia dose: The test first screens the appropriate dose range of dexmedetomidine combined with zoletil, four dose groups were set up,0.1 mg/kg dexmedetomidine combined with 6.25 mg/kg zoletil,0.1 mg/kg dexmedetomidine combined with 7.5 mg/kg zoletil,0.15 mg/kg dexmedetomidine combined with 6.25 mg/kg zoletil,0.15mg/kg dexmedetomidine combined with 7.5 mg/kg zoletil.A total of eight groups were tested in two ways of oral administration, direct oral administration and oral gel, with six experimental cats in each group. The time of anesthesia induction period, maintenance period and recovery period was recorded, and the sedative, analgesic, and muscle relaxation effects at different time points were scored, and those with the best effect were screened out.(3)Anesthetize the experimental cat with the screening dose, and the basic physiological indexes of the experimental cats before anesthesia,1h during anesthesia and 24 h after anesthesia were monitored, and the blood routine, blood biochemistry, arterial blood gas and other indexes were detected.Comprehensive evaluation the safety of screening doses during oral anesthesia in cats.(4)The clinical application test was carried out with the screening dose of two oral anesthesia methods, and the number of cats that successfully took it,produced anesthesia effect, produced braking effect was recorded.Test results:(1)Among the various anesthetic formulations, dexmedetomidine combined with Zoletil direct oral anesthesia has suitable anesthesia induction time, stable recovery, and the anesthetic prescriptions with less side effects on cats.(2)Among the test doses, the direct oral administration group of0.1 mg/kg dexmedetomidine combined with 7.5 mg/kg Zoletil had the best anesthesia effect.Anesthesia induction in this group was stable, with an average of 30.00 ± 3.40 min.The mean duration of anesthesia was 74.00 ± 4.73 min,and the anesthesia effect is stable; The oral gel group also had the best anesthesia effect of 0.1 mg/kg dexmedetomidine combined with 7.5 mg/kg Zoletil, but compared with the oral administration group, the oral gel group induced anesthesia more quickly, with an average anesthesia induction time of 21.67 ± 2.58 min,and then gradually wakes up.The anesthesia score is not higher than 7in the whole process, can provide about 44.83 ± 3.13 min of anesthesia.(3)At the screening dose, the blood oxygen saturation of the experimental cat was not lower than 95% during anesthesia. The anesthesia prescription had little effect on the cat’s circulation and respiratory system, and no obvious side effects were found.Laboratory test results: The changes in the blood routine, blood biochemistry, arterial blood gas and other indexes of the experimental cat were all within a reasonable range, and the blood indexes after 24 h were not significantly different from those before the test(p < 0.05).(4)At the screening dose, nine out of ten stray cats in the direct oral administration group successfully took anesthetic drugs,of which eight cats entered anesthesia, one cat had a good braking effect.Seven of the ten pet cats in the oral gel group successfully took anesthetics,of which six cats had anesthesia effect, one cat had a braking effect.Based on the above test results,it is concluded that:(1)Dexmedetomidine 0.1 mg/kg combined with Zoletil 7.5 mg/kg dircet oral anesthesia have stable induction effects, the anesthesia maintenance time was longer, the anesthesia state was deeper during the maintenance period, and the side effects were small.(2)Dexmedetomidine 0.1 mg/kg combined with Zoletil 7.5 mg/kg oral gel anesthesia is induced rapidly, the duration and the depth of anesthesia were appropriate, and the recovery period was stable.(3)The two kinds of oral anesthesia have short-lived and recoverable effects on the basic physiological functions of cats, and have no significant effect on the blood routine a,blood gas,and the biochemical functions of the liver and kidney.(4)Two oral anesthesia methods are applied to cats, with strong acceptability, high success rate of administration, stable anesthesia effect and high clinical practicability.
