| Rabbits play an important role in experimental animals,and because of their timid and sensitive characteristics,anesthesia is very important in experiments and operations.However,there are no special anesthetics for rabbits at home and abroad,and the existing anesthetics have their own limitations in rabbit anesthesia.Current studies have shown that the combination of two or more drugs can achieve better anesthesia results and improve the safety of anesthesia compared to the use of only one drug.The combination of Zoletil and Dexmedetomidine can produce synergistic effect,enhance the depth of anesthesia,reduce the dosage,and reduce the risk of anesthesia.In order to verify the anesthesia effect of Zoletil combined with Dexmedetomidine on rabbits,different doses of Zoletil combined with Dexmedetomidine were used in the anesthesia of rabbits in this study,and various physiological indicators in the anesthesia process were observed,so as to provide certain reference for finding safe and effective anesthesia methods suitable for rabbits.Twenty-five experimental rabbits were randomly divided into 5 groups with 5rabbits in each group.The 5 experimental groups were given combined anesthesia with Zoletil and Dexmedetomidine,and were divided into Dexmedetomidine dose fixed group(Group AX)and Zoletil dose fixed group(Group BX).The drug doses for each group were: Group A1(low dose): Zoletil 2.5 mg/kg,Dexmedetomidine 12.5 μg/kg;Group A2(medium dose): Zoletil 5 mg/kg,Dexmedetomidine 12.5 μg/kg;Group A3(high dose): Zoletil 7.5 mg/kg,Dexmedetomidine 12.5 μg/kg;Group B1(medium dose):Zoletil 2.5 mg/kg,Dexmedetomidine 25 μg/kg;Group B2(high dose): Zoletil 2.5 mg/kg,Dexmedetomidine 37.5 μg/kg.All groups were injected intravenously with Zoletil first,and then intramuscularly injected with Dexmedetomidine immediately after completion,and the time of completion was recorded as 0 min,and the time of anesthesia stages(induction period,anesthesia period,recovery period)was recorded.Physiological indexes(heart rate,respiratory rate,blood oxygen saturation,body temperature),biological reflex activity(anal reflex,eyelid reflex,corneal reflex)and anesthesia depth(sedation,analgesia,muscle relaxation)of rabbits were recorded before anesthesia,at 5min,10 min,20 min,30 min,40 min,50 min and 60 min after anesthesia.According to the above data analysis,the optimal dose was screened out,and median sternotomy and gastrotomy were performed after the rabbits were anesthetized with this dose to conduct surgical verification tests to evaluate the clinical applicability of the anesthesia method.The experimental results showed that the induction time of each group was about17 s.Anesthesia time: Group A1: 22.88±3.93 min;Group A2: 50.99±16.08 min;Group A3: 66.07±11.52 min;Group B1: 57.57±9.71 min;Group B2: 64.73±15.17 min.The awakening time of A1 group was 29.49±6.45 min,and that of other groups was approximately equal,about 54 min.The effective anesthesia time(the time for maintaining the anesthesia depth suitable for operation)was shorter in A1 and A2 groups,about 40 min in B1 group and 45 min in A3 and B2 groups.During anesthesia monitoring,the heart rate of each group increased first and then decreased,and the heart rate of B1 and B2 groups decreased more obviously;the body temperature of each group kept decreasing,and the decrease was equal;the respiratory rate and blood oxygen saturation of each group decreased first and then rebounded,and the changes were within the physiological tolerance range;the reflex activity of each group was inhibited to different degrees,and the depth of anesthesia of A3,B1 and B2 groups was better than that of A1 and A2 groups.Some animals(2/5)in A3 group showed agitation during the recovery period,while no obvious abnormal symptoms were observed in other groups during anesthesia.All the above changes are recovered within a certain period of time.The optimal dose finally screened out in this trial is B1 group: Zoletil 2.5 mg/kg and Dexmedetomidine 25 μg/kg.Applying this dose to a surgical validation trial in rabbits,the results showed that the induction period of anesthesia was short,the anesthesia time reached 57.34±4.61 min,and the operation was completed smoothly during the anesthesia period.The rabbits recovered smoothly after the operation,and the physiological indexes recovered in a certain period of time,and no obvious adverse reactions were found.The results showed that the combined anesthesia of Zoletil and Dexmedetomidine could achieve good anesthesia effect in rabbits,and there is a significant dose dependence in low dose to medium dose.Intravenous injection of Zoletil 2.5 mg/kg in rabbits,followed by immediate intramuscular injection of Dexmedetomidine 25 μg/kg,anesthesia induction is rapid,anesthesia time reached 57.57±9.71 min,sedation,analgesia,muscle relaxation effect is good,and high safety.To meet the needs of clinical surgery and experimental operations. |
