Objective: To observe the effect of Yang-yin-huo-wei Mixture on CAG rats gastric mucosal apoptosis cells, gastric mucosa Bcl-2 protein and TFF1 mRNA and protein content, and to elaborated mechanism of Yang-yin-huo-wei Mixture in the treatment of CAG. Methods: The rats were randomly divided into blank control group, model group, low dose of Chinese medicine group, middle dose group, high dose group, positive drug group, 16 rats in each group. In addition to the blank control group conventional farming, free access to food and water, The other 5 group using 2g sodium salicylate added 100 mL of 30% ethanol to rats by gavage,1 times a day, each time 3mL, with the Fed on alternate days not banned water caused by rat model of chronic atrophic gastritis, model successfully reproduced in traditional Chinese medicine in the treatment of high, in the low dose group, respectively,feed Yang-yin-huo-wei Mixture of water decoction of 2.22g/kg/D,1.48g/kg/D,and0.74g/kg/D. After 24 weeks, to observation gastric mucosa morphology, determination of each rat gastric mucosa Bcl-2 protein,TFF1 mRNA and protein content, TUNEL determination of traditional Chinese medicine treatment of each rat cell apoptosis change. Results: Compared with the blank control group, the rats in the model group AI decreased significantly, TFF1 protein and mRNA content decreased. In traditional Chinese medicine,low dose group and positive drug group TFF1 protein significantly decreased, Bcl-2 protein in all groups increased in varying degrees, the difference was statistically significant(P<0.05), proving that copy the successful model. Compared with the model group,high dosage, middle dose and positive drug group of TFF1 protein and TFF1 mRNA content were significantly increased, Bcl-2 protein significantly decreased, the difference was statistically significant(P<0.05); traditional Chinese medicine group and positive drug group AI significantly increased,the difference was statistically significant(P<0.05). Compared with the positive drug group, high dose of Chinese medicine group, AI, TFF1 protein and mRNA content were significantly increased, Bcl-2 protein decreased significantly, the difference was statistically significant(P<0.05).Compared with the low dose group, AI, TFF1 protein and mRNA content were significantly increased, Bcl-2 protein was significantly lower, the difference was statistically significant(P<0.05).Conclusion:(1)Yang-yin-huo-wei Mixture can by down regulation of Bcl-2 protein level and up regulation of the TFF1 protein and mRNA content, so as to achieve the purpose of treatment of CAG.(2)Yang-yin-huo-wei Mixture can promote cell apoptosis, and prompted abnormal proliferation of gastric mucosa cells apoptosis contributes to the improvement of the CAG.
