Objective:To observe the effect of Yang-yin-huo-wei Mixture on CAG rats’gastric mucosal cells and its TLRs signal transduction pathway. And to revealed Yang-yin-huo-wei Mixture the treatment ways and targets in treating CAG. Elaborated the separately mechanism of Yang-yin-huo-wei Mixture in the treatment of CAG from the molecμlar level and protein level,and to observe the anti inflammation changes by determination inflammatory factors. Methods:Randomly divide the experimental animal into 6 groups:blank control group, model group, positive drug group, low dose of Chinese medicine group, middle dose group, high dose group,16 rats in each group. Except blank control group conventional breeding, free access to food and water,30% ethanol solution and the remaining 5 groups 2g sodium salicylate adding 100 mL, Ig rats,1 times a day, each time 3 mL, with the Fed on alternate days coμldn’t water the establishment of atrophic gastritis rat model, model successfμlly reproduced in traditional Chinese medicine in the treatment of low, in the high dose group, respectively, feed Yang-yin-huo-wei Mixture of water decoction of 0.74g/kg/D,1.48g/kg/D,and2.22g/kg/d. After 12 weeks in the determination of serum IL-6, TNF-α, IFN-β and NOS2 inflammatory factor levels, measure the original TLRs and downstream transduction mRNA and protein content.Resμlts:Yang-yin-huo-wei Mixture can improve the rat gastric mucosa pathological state, lead to the normal gastric mucosa lesion. Compared with blank control group, each group inflammatory factor and TIRs and downstream signal transduction proteins and mRNA content were significantly increased (P<0.05), proving that copy the successfμl model. Compared with model group, high dose group of TNF-α, IL-6, NOS2 levels were significantly lower, positive drug control group and the traditional Chinese medicine dose group of TIRs and downstream signal transduction proteins and mRNA content reduced to some extent, the difference was statistically significant (P<0.05). And the comparison between Yin "stomach dose group, Chinese medicine IFN-P lower dose group, high dose group TIRs and downstream signal transduction proteins and the decline in mRNA content to some extent, the difference was statistically significant(P<0.05). Conclusion:(1) Yang-yin-huo-wei Mixture can live original sample TLR receptors and their downstream transduction genes and protein level, reduce the release of inflammatory factor by TLRs pathway activation of gastric mucosal damage, so as to achieve the purpose of the treatment of chronic atrophic gastritis. (2) Yang-yin-huo-wei Mixture can reduce chronic atrophic gastritis rats serum level of inflammatory factors, and inhibit the inflammatory response may contribute to the improvement of chronic atrophic gastritis disease.
