The Research Of ManWeiLing(MWL) Mixture Treatment On Rats With Chronic Atrophic Gastrictis

Objective: To observe the effect of ManWeiLing(MWL) mixture on the pepsin activity and Gastric juice PH and the serum level of SOD,MDA and the Proliferating cell nuclear antigen and gastric mucosa pathological histology in rats model of Chronic Atrophic Gastritis, and to explore the mechanism in the pretreatment of Chronic Atrophic Gastritis.Methods: Sixty Wistar rats were randomly divided into five groups: normal control group,model group,low-dose MWL group,middle-dose MWL group and high-dose MWL group. Except normal control group rats, CAG model was established in other rats by synthetically method:2%salicylas were irrigated into the stomach of the rats once a day, and 0.02%hartshorns was drink freely et al, all the factors were given for two months. when the model was established successfully, model group rats were put to death, their Gastric juice pH were determined, their blood and stomachs were preserved, other rats were cured with low–dose MWL,middle-dose MWL and high-dose MWL. After six weeks, all the rats were put to death, their Gastric juice pH were determined,their blood and stomachs were preserved to detect the serum level of SOD,MDA and the chymase activity and observe the histology mucosa changes by microscopy. Their stomachs were to detect PCNA by immunohistochemistry methods.Results: Compared to normal control group, the Gastric juice pH in the rats of model group was significantly higher(P<0.01),and the pepsin activity was significantly lower(P<0.01)and the serum level of SOD was significantly lower(P<0.01),MDA was significantly higher(P<0.01)and the PCNA was significantly higher(P<0.01).The pathologic changesof gastric mucosa were significantly worse. Those rats were cured with different dos medicines, for six weeks, there was significant statistics difference in pathologic changes of gastric mucosa,Gastric juice pH,pepsin activity,SOD,MDA and PCNA from model group(P<0.01). low–dose MWL group,middle-dose MWL group and high-dose MWL group also have different significant statistics. (P<0.01 or P<0.05).Conclusion: The higher Gastric juice pH,SOD and lower pepsin activity,MDA and the expression of PCNA are important display of CAG. There is an important effect to heighten the pepsin activity and reduce the Gastric juice pH for CAG after being cured by MWL. MWL could scavenge significantly free radical and have protective functions against the gastric mucosal injure and inhibit the abnormal proliferating of rats gastric mucosa cell, even can turnover partly the pathologic changes of gastric mucosa. With the dose of MWL increasing, the curative result will be more remarkable.
