Objective: With the prepared traditional Chinese medicine hollow bolt suppository(Fufangdiyu hollow type suppository,which is referred to as FFDYHS) acting on the rat model of perimenopausal syndrome and related testing indicators, to explore the effects and mechanisms for FFDYHS on perimenopausal syndrome from the molecular level, in order to further lay the foundation for the research and the use of clinic.Methods: 1 To ground FFDYHS prescription ShuDi,ShanZhuyu,HuangLian,Gou Teng,Shan Yao,Fu Shen,Yuan Zhi, Xiang Fu, Dan Shen, Ge Gen, Bai He, Fu Xiaomai,Gan Cao, total of 13 herbs in a mortar, through a 120 mesh sieve to get compound free fried particles powder, prepared by a specific method of literature tutor previous study and the associated determined experimentally prepared hollow type suppositor for rats each piece weighs about 1.905 g, free fried particles containing the compound dose 0.333 g. 1 Selected 48 female students of childbearing age which are not pregnant SD rats randomly divided into sham-operated control group, model group,positive group, Fufang fed group, the high dose group of FFDYHS, the low dose group of FFDYHS. In addition to the skin incision was sutured back immediately the sham control group incision after a small amount of adipose tissue, the remaining 5groups of rats remove both ovaries, and menopausal syndrome rat model was prepared. Rats were fed a week of postoperative recovery groups, and three days in a row before the intramuscular injection of penicillin to 40,000 units, 1 time/day, to prevent infection. A week later, the rats were observed vaginal cytology, 1 time/day for five consecutive days, excluding(modeling unsuccessful) rats whose castration is not complete. 1After vaginal smears,the sham control group and model group were given blank blank hollow type suppository, positive group were given estradiol valerate and Fufang fed group were given Di Yu Fang decoction,FFDYHS high-dose group and low-dose groups, respectively given the high and low dose FFDYHS.Continuous administration of 14 days, 24 hours after the last administration ready to kill the subject, the subject 12 hours before the killing can not help but water fasting,abdominal aortic blood, quickly decapitated rats, the removal of the hypothalamus.Serum estradiol, follicle stimulating hormone, luteinizing hormone and hypothalamusβ-EP by Elisa kits.Result: 1FFDYHS Can increase serum E2 levels in rats while reducing FSH, LH levels, compared with model group were significantly different(P<0.05), Compared with positive group and oral medicine group had no significant difference(P>0.05).1FFDYHS Can increase hypothalamic β-EP, compared with the model group were significantly different(P<0.05), oral medicine group had no significant difference(P>0.05).Conclusion:FFDYHS can regulate the secretion of hormone levels and hypothalamicβ-EP of the rats of perimenopausal syndrome.so E2 and β-EP increased, FSH and LH decreased, thereby improving the clinical symptoms of climacteric syndrome. The mechanism may be involved in the regulation of neuroendocrine system perimenopausal syndrome by regulating the function of these indicators directly or indirectly.
