Purpose:This study observes the effects of Guipi Decoction on the IL-1βand expression of IL-1 receptorⅠ,to discuss the anti-depression mechanism of Guipi Decoction.Material and method:①Forty common female Wistar rats were selected,which having divided into 4 groups:the normal group,the model group,the saline group and the Chinese medicine group with 10 rats per group.②Except the normal group, rats in other 3 groups were established the depression model by received chronic unpredictable stress stimulus.③No medicine was given to the normal group.The model group was exerted with gavaged action.The saline group was gavaged with 3ml saline 0.5h before the stimulus.The Chinese medicine group was gavaged with 3ml Guipi Decoction at the same time.④All rats were observed weight and taken Open-field test on the 0 and 22nd day.⑤Fluorescent dye to detect the rats hippocampal cell.⑥Macrophage phagocytosis test to detect the rats immunity function.⑦The serum IL-1βwas detected with radio-immunity.⑧The expression of IL-1RⅠwas detected with immunohistochemical method.The expression cells number,optical density and gray degree of IL-1RⅠwas measured with BI-2000 medical image analysis system.⑨The data was expressed by(?)±s,statistical analyzing with SPSS11.0.Results:1 Weight of each group:Compared with the normal group,the weight of model group and saline group decreased obviously(P<0.01),there was no difference between the Chinese medicine group and the normal group.2.Open-field test:Compared with the normal group,the behavior points of the model group and saline group reduced obviously(P<0.01),and there was no difference between the Chinese medicine group and the normal group.3.Hippocampal cell:Observed under fluorescence microscopy,compared with the normal group,the model group and the saline group hippocampal cells were not completed,chromatic agglutination,cells nuclei from shrinkage,condensation and cleavage showed that the cells apoptosis.Necrotic cells showed red fluorescence.4.Phagocytic index:Observed under oil immersion lens,compared with the normal group,the model group and the saline group reduced obviously(P<0.05),and there was no difference between the Chinese medicine group and the normal group.5.The serum IL-1β:Compared with the normal group,the model group and the saline group increased obviously(P<0.01),and there was no difference between the Chinese medicine group and the normal group.6.The expression of IL-1RⅠ:Compared with the normal group,the model group and the saline group increased obviously(P<0.05),and there was no difference between the Chinese medicine group and the normal group.Conclusion:1.Guipi Decoction could protect the hippocampal cells from chronic stress.2.Guipi Decoction could maintain the normal phagocytic index in chronic unpredictable stress.3.Guipi Decoction could antagonize increasing the excretion of IL-1βby chronic unpredictable stress.4.Guipi Decoction could antagonize increasing IL-1 RⅠexpression by chronic unpredictable stress.
