Purpose:To study the effect of Guipi Decoction on rats' hippocampal NGF and its receptor expression,serum T4 concentration,preliminary study its possible control mechanism of depression.Material and method:①The 60 healthy adult female Wistar rats reared adaptive one-week,40 rats were randomly selected,which having the similar weight,behavior points were divided into the normal group,model group,saline group and Chinese medicine group.With 10 rats in each group.②Except the normal group,rats in the other 3 group were established the depression model by received unpredictable stress stimulus.③control group was not administered;model group action only imposed but not oral garage;saline group 2ml normal saline garage;Chinese Medicines garage 2ml Guipi Decoction.Health and dosage used under adult dosage of the drug in rats was converted to the amount of enrichment after decoction made to 2ml.Above each group were half an hour before each stimulus imposed administered orally or action.④At 1 days and 21 days measure the body weight and then use open-field method to measure behavioral score.⑤SABC immunohistochemical assay hippocampal neurons cells NGF content and its receptor expression changes,BI-2000 Medical Image Analysis System for counting the number of immunopositive cells and scan gray value.⑥using PEG double-antibody radioimmunoassay method for determination of serum T4 level of rats.⑦data to(?)±S said that the use of statistical software to conduct statistical SPSS10.0 treatment.Results:①The body weight of model group and the saline group rats,compared with the control group,decreased,.there is statistical significance(p<0.05); Chinese medicine group and the control group no significant difference.②Model group and the saline group of rats behavioral score compared with the control group were significantly decreased,there is statistical significance(p<0.01); Chinese medicine group and the control group no significant difference.③Immunohistochemical method to detect,model group and saline group rat hippocampal neurons intracellular expression of NGF and its receptor content decreased compared with the control group there is significant difference(p<0.01),the Chinese Medicines Board did not change significantly.④PEG double antibody RIA measured,model group and the saline content of serum T4 was lower than the control group(p<0.05),traditional Chinese medicine group and the control group compared to no significant change.Conclusion:①Guipi Decoction can protect hippocampal neurons against chronic unpredictable stress damage.②Guipi Decoction can improve depression model rats' hippocampal neurons with NGF and its receptor expression in the content, thereby protecting hippocampal neurons.③Guipi Decoction can improve serum T4 level of depression model rats.④The antidepressant mechanisms of Guipi Decoction are possible by improving the depression model rats' hippocampal neurons NGF and its receptor express and serum T4 level.
