PartⅠ: The establishment of a model of the intervertebral disc degeneration of Passive Cigarett-Smoking of RatsObjective: To observe the effect of passive smoking to intervertebral disc of rats.Methods: 60 4-week-old SD rats were randomly divided into 6 groups averagely, the 6 groups were treated with non-smoking as control (group 1), with 2,4,8 weeks of passive cigarette-smoking (group 2,3,4), with 4 and 8 weeks of cessation of passive cigarette-smoking after 8 weeks of passive cigarette-smoking (group 5,6). L4~L5 intervertebral disc were excised after the rats were killed .HE staining was used to check the histological changes.Results: There were not clear change in intervertebral discs of group 2 from HE staining, group 3,4'degeneration reached grade 2~3 and grade 3~4 respectively. The disc degeneration were repaired to some degree after 8-week but no after 4-week cessation of smoking.Conclusion: Passive smoking can reduce intervertebral disc degeneration, the degree of degeneration was related with the duration of passive smoking PartⅡ: Expression of interleukin-1β,interleukin-1 receptorⅠand their significance in the intervertebral disc degeneration of passive Cigarett-Smoking in ratsObjective: To investigate the expression of expression of interleukin-1β,interleukin-1 receptorⅠafter establishment of a model of the intervertebral disc degeneration of Passive Cigarett-Smoking of rats.Methods: After establishment of a model of the intervertebral disc degeneration of Passive Cigarett-Smoking of Rats, IL-1β,IL-1 receptorⅠwas examined by using immunohistochemical SP staining.Results: The rates of IL-1β,IL-1 receptorⅠpositive-staining cells rised, and reached 76.3±3.24 and 46.3±2.79 respectively after 8-week of passive smoking, then decreased after 4-week cessation of smoking, and dropped to 66.±2.94% and 38.±2.23% respectively.Conclusion: Passive smoking can reduce irreversible intervertebral disc degeneration, and Interleukin-1β,Interleukin-1 receptorⅠmaybe are the important factors in aggravation of degeneration.
