Objective: To study the anti-depression effect of Guipi Decoction and the mechanism is approached.Method: (1)40 common female Wistar rats were selected, which having the same weight, behavior points and learning memory ability. They were divided into the control group, the model group, the saline group and the Chinese medicine group with 10 rats per group. (2)Except the control group, rats in other 3 group were established the depression model by received chronic unpredictable stress stimulus. (3)No medicine was given to the control group. The model group was exerted with lavage action. The saline group was lavaged with 2ml saline. The Chinese medicine group was lavaged with Guipi Decoction 0.5h earlier the stimulus. (4)The body weight was measured with GS-671 electronic scale. (5)The behavior points were measured with open field method. (6)The learning memory ability was measured with Morris water maze video tracking system. (7)The histology changes of CA3 neurons were observed with HE staining. Neural number was measured with BI-2000 medical image analysis system. (8)The serum cortisol was detected with ELISA. (9)The data was expressed by x ± s , statistical analyzing with SPSS 10.0.Result: (1)Compared with the control group, the body weight of model group and saline group decreased obviously, which was significant in statistics. But body weight of Chinese medicine group did not change much compared with the control group. (2)Compared with the control group, the behavior points of the model group and saline group reduced obviously, and those of the Chinese medicine did not change significantly. (3)Compared with the control group, the escape latent time and the swimming distances of the model group and saline group increased obviously, which was valuable in statistics. But the two indexes of the Chinese group did not change significantly compared with the control group. (4)In the CA3 areas, there were abundant pyramidal cell layers, and cells were complete, compact, well-arranged, and the edge of cells was clear in the control and Chinese medicine groups. While the layers of pyramidal cell decreased, and the cleft of cell enlarged, and the array of cell was sparse, even many cells were deficient in the model and saline groups. Compared with the control group, the neuron number of the model and saline groups decreased obviously and that of the Chinese medicine group did not change significantly. (5)Compared with the control group, serum cortisol concentrations in the model group and saline group increased obviously, which was significant in statistics. And no obvious changes existed in the control group and the Chinese medicine group.Conclusion: (1)Guipi Decoction might antagonize the decreasing of rat weight, behavior points and learning memory ability caused by chronic unpredictable stress. (3)Guipi Decoction might protect the hippocampal CA3 neurons from the injuries by chronic unpredictable stress. (3)Guipi Decoction might resist the supersecretion of cortisol by chronic unpredictable stress.
