Studies on the prevention and cure of the contamination during plant tissue culture were conducted in Agricultural University of Shanxi during 2003 ~2005, using seven kinds of plant(Rosa chinensis , Exbucklandia populnea, Rubus spp. ,Juglans regia,Prunus persica,Populus tomentosa ,Ziziphus jujube)as trial materials. The main results were as follows:1. The explants of three kinds plants were tested of contamination. The experiment showed that the rate of contamination are, correspondingly ,more than 95%(stem of Exbucklandia populnea), 65.4%( a part of stem of Juglans regia),less than 5%( auxiliary bud of Populus tomentosa).2. There are higher rate of contamination that explants were grown in field compare with grown in hothouse.especially for Prunus persica.Their disparity were very obvious,11%(Prunus persica),8%(Populus tomentosa),6%( Ziziphus jujube) on former and latter.3. The top branches and annual sprouts of Prunus persica were inoculated separately in March, August and November. In accordance with the rate of contamination and start ,March is best season to inoculate for the top branches of Prunus persica.4. Trial materials (Prunus persica) were pretreated with diluted 2000times 70% thiophanate-methyl, the survival rate of inoculation has risen greatly.5. Three kinds levels on mixed bactericides composed by Sulphur, Penicillin, Tetracycline, Hormone and PPA were used in the test to control the culture shoots. Trial materials were soaked in those drugs for 24h,the survival rate come up to 85%(Populus tomentosa)and 87%(Prunus persica).6. The small stem tuber of Exbucklandia populnea were treated with hot water (51 li),which not only reduce the rate of contamination but also less be less harmful to explants.7. Those contaminated clustering shoots of Rubus spp. were treated with different kinds and concentration antiseptic , which 2% Sorbicacid was best drug that control time longer.8. The Results were best that the clustering shoots of Rubus spp. were treated with 250 ppm Ampicillin Sodium.9. Accrding to colony character of the microbe , taking photograph by microscope, shape character, the identification is made. The contamination fungus are and bacterium are Trihoderma, Penicillium sp., Aspergillus niger, Rhizoctonia, Actinomyces, Alternaria.
