In the process of plant tissue culture,the explant(cell clusters,adventitious buds or plantlets,etc.)smaller than the growth rate of microorganisms,long production cycle,vulnerable to pollution,which seriously affects the economic benefits of plant tissue culture.In this study,we analyzed the pollution in plant tissue culture seedling factory,and select the most kinds of bacteria,and 4 strains were obtained by the tip of the mycelium.From the morphological point of view,the strains of Fusarium were identified.The results showed that the 4 strains were Fusarium Penicillium,Aspergillus niger,and Bacillus pumilus,respectively.In order to control pollution and reduce production cost effectively,their growth rate and growth curve were investigated.the production process parameters and the concentration of nano-silver particles were discussed:1.The optimum reaction conditions were: the concentration of silver nitrate and sodium nitrate was higher than that of 3.85:1,the concentration of silver nitrate and sodium nitrate was higher than that of 0.22mmol/L,and the reaction time was three days.2.The identification show that Penicillium,Fusarium,Aspergillus niger and Bacillus pumilus strain are the main cause of contamination of plant tissue culture.3.Adding nano-silver antibacterial medium for tissue culture process of the 4 major pollution bacteria have obvious effect of killing,in the 0.5 mg/L-4.0mg/L concentration range,with the increase of nano-silver concentration,antibacterial ability gradually increased,the concentration of 5mg/L can reduce the existing pollution rate of 75%,has practical application significance.
