Compound anesthesia is an anesthetic technique that uses two or more anesthetic drugs and anesthetic methods for clinical experiments, with the aim of obtaining satisfactory intraoperative anesthetic and postoperative analgesic effects and providing a good environment for animal surgery. In recent years, the development and application of compound anesthesia has gradually become a new trend in the development of animal medical anesthesia. As an imported injectable anesthetic drug commonly used in animal clinics,Zoletil has a rapid induction process, good muscle relaxation effect and high safety factor, but the disadvantages are short duration of anesthesia, easy to lead to increased airway secretions thus leading to airway obstruction breathing control or pulmonary edema, vomiting, tachycardia and other reactions during the induction period of anesthesia. The other anesthetic drug Dexmedetomidine Hydrochloride, through a large number of clinical experiments,it has been concluded that it has little side effects on animals during anesthesia and almost no adverse reactions, and because of the good analgesic and sedative effect,it can be used in combination with other anesthetics to achieve better clinical anesthetic effects. Therefore,in this experiment, the combination of Zoletil and Dexmedetomidine Hydrochloride was used for the combination of anesthesia in small-tailed cold sheep, and different monitoring methods were used to make a comprehensive analysis of the test animal organism and the anesthesia method used.In this trial,15 clinically healthy and immunized small-tailed cold sheep were randomly selected and divided into 5 groups of 3 sheep each. Each sheep in group I was given 0.05 m L/kg(2.5 mg/kg)of Zoletil intravenously and 0.02 m L/kg(5 μg/kg)of Dexmedetomidine Hydrochloride intramuscularly;each sheep in group II was given 0.05 m L/kg(2.5 mg/kg)of Zoletil intravenously and 0.06 m L/kg(15(15 μg/kg)of Dexmedetomidine Hydrochloride;compound group III: 0.05 m L/kg(2.5 mg/kg)of Zoletil intravenously and 0.04 m L/kg(10 μg/kg)of Dexmedetomidine Hydrochloride intramuscularly per sheep;compound group IV: 0.075 m L/kg(3.75 mg/kg)of Zoletil intravenously per sheep and 0.075 m L/kg(3.75 mg/kg)of Dexmedetomidine Hydrochloride intramuscularly per sheep. In Group IV,each sheep was given 0.075 m L/kg(3.75 mg/kg)of Zoletil intravenously and 0.04 m L/kg(10 μg/kg)of Dexmedetomidine Hydrochloride intramuscularly;in Group V,each sheep was given0.025 m L/kg(1.25 mg/kg)of Zoletil intravenously and 0.04 m L/kg(10 μg/kg)of Dexmedetomidine Hydrochloride intramuscularly. After that, the physiological indexes(heart rate, respiratory rate, body temperature, oxygen saturation),reflex activity(anal reflex, eyelid reflex, corneal reflex),and reflexes were recorded for 5min,10 min,20 min,30 min,40 min,50 min,60 min,70 min,80 min,and 90 min for each sheep respectively.reflexes, corneal reflexes)and anesthetic effects(sedation, analgesia, muscle relaxation)as well as their natural awakening time. The results of anesthesia time and analgesia time tests in each group were101.58±10.70 min and 37.63±4.83 min in compound group I;158.64±8.15 min and94.23±2.36 min in compound group II;152.09±28.72 min and 86.46±8.45 min in compound group III;and 179.46±22.58 min in compound group IV.There was no reflex activity during the anesthetic analgesic period, and the anesthetic effect was good. The best anesthesia compound solution group selected in this experiment was compound group III,and its anesthetic dose and method(0.05 m L/kg(2.5 mg/kg)of intravenous injection of Zoletil and 0.04 m L/kg(10 μg/kg)of intramuscular injection of Dexmedetomidine Hydrochloride)were applied to the clinical diagnosis of a case of a small-tailed cold sheep that had accumulated too much food and could not continue eating. In the treatment, the data monitoring values and the anesthetic effect during the surgical anesthesia were well performed and were basically consistent with the monitoring data of the previous trial.Collectively, the use of Zoletil or Dexmedetomidine Hydrochloride for compound anesthesia in animal anesthesia was more effective, had fewer adverse effects, performed better for postoperative recovery and activities than the known single anesthesia using only Zoletil or Dexmedetomidine Hydrochloride. In the experiment, the compound anesthetic regimen of 0.05 m L/kg(2.5 mg/kg)of Zoletil intravenously and 0.04 m L/kg(10 μg/kg)of Dexmedetomidine Hydrochloride intramuscularly in the compound III group showed better anesthetic effect, higher safety and lower risk factor compared with other groups, and this dose was the optimal regimen in the tested. This compound anesthesia dose and method provides a selective and exact scientific theoretical basis for clinical anesthesia not only for sheep but also for other large animals.
