Oral anesthesia is a less studied anesthetic method in veterinary field,but in human medicine,the oral administration of anesthetics is very important,and it can be recognized by patients or their families in clinical application.Oral administration can reduce the vigilance of animals and avoid the stress reaction of animals in the course of administration.It has the characteristics of convenient operation and wide safety range.It can guarantee the safety of the anesthetic during the anaesthesia.Oral anesthesia is of great significance in animal capture,long-distance transportation and wildlife anesthesia.In this experiment,?-chloraldose was taken orally as the object of study,combine with Dexmedetomidine to anaesthesia dogs.To select the best anesthetic formula for oral administration of this compound preparation in dogs,and verify the feasibility and safety of this anesthesia methods.Provide new research ideas and experimental basis for animal anaesthesia.Test content:(1)Screening of anesthetic prescription: The trial was divided into five groups according to the dose gradient.Canning of fluids mixed with different concentrations of compound anesthetic agents in dogs.Monitoring the basic physiological indexes of dogs at different time points.Record the duration of induction,maintenance and recovery of anesthesia,and score the anesthesia to select the optimal group.(2)The optimal formula was used for laboratory test,including the blood routine index,blood biochemical index and arterial blood gas of 24 h and 48 h after the experiment,before and after the experiment,and the change of the non-invasive blood pressure during the anesthesia of the experimental dog.Verify that the formula will cause organ damage or blood index effects on the animal body.(3)Determination of anesthetic index: the half-lethal lethal dose(LD50)and the half effective dose(ED50)of the group were determined by a mouse experiment.Calculate the anesthetic index of the composite reagent,and the safety was evaluated.Test results:(1)Among the experimental doses,360 mg/kg ?-chloralose complex 0.6 mg/kg dexmedetomidine had the best anesthesia.This group has the shortest induction time for anesthesia,the time is 13.83 ± 2.93 min.The duration of anesthesia maintenance period is relatively long in other groups,he time is 67.83 ± 8.40 min.The anesthetic effect is stable.The anesthesia score can be maintained > 7 within 20-80 min after taking the medicine.The basic physiological index of the body is less affected than the high dose group,and the heart rate is reduced to 58.83 ± 3.13beats/min,and the respiratory rate is reduced to the lowest 7.00 ± 1.09 beats/min,the body temperature drops to 36.65 ± 0.21°C;(2)Laboratory test results: All the indexes of theexperimental dogs did not change significantly beyond the normal range,only the p H changed significantly,but still in the normal range.After 48 hours,the blood indexes returned to normal.There was no significant difference between the two groups before the experiment.There was no significant change in non-invasive blood pressure during anesthesia.(3)Measurement result of anesthesia index: The median lethal dose(LD50)of the oral composite preparation was calculated to be 157.85 mg/kg,and the half effective amount was(ED50)28.12 mg/kg,and the anesthetic index(AI)was 5.61.Based on the above test results:(1)?-chloraldose 360 mg/kg combined with dexmedetomidine 0.6 mg/kg had better induction effect and less side effect.(2)The composite preparation did not cause significant changes in blood parameters of the animal body,and did not cause damage to the liver function and renal function of the body.The anesthetic index was 5.61 >3,which was safe and could satisfy the need of superficial surgery and animal capture and transportation.
