Effect Of Bailing Capsule On Expression Of Autophagy Related Protein LC3 And Beclin-1 In Renal Tissue Of Rats After Renal Impact Injury

Objective:In this study,we studied the effect of Bailing Capsule on the expression of autophagy related protein LC3 and Beclin-1 in rat kidney after blunt impact injury.Methods:A total of 30 healthy male SD rats were randomly divided into three groups:(1)normal control group(group A,n=10);(2)Kidney impingement injury model group(B group,n=10);(3)Bailing Capsule group(group C,n=10).After anesthesia,rats in group B and C were given BIM-V biological impact machine to establish blunt impact injury model of kidney.After impact modeling,rats in group C were given bailing capsule dissolved in normal saline by gavage(1.5g/(Kg·d)、3 times/day).Group B was given equal amount of normal saline after the impact of the model making,and group A did not impact after anesthesia.Rats in group A,group B and group C were given continuous gavage for 2 weeks,and blood was collected through the posterior orbital vein plexus and the kidney was taken out.The pathological changes of kidney tissue were observed by hematoxylin-eosin(HE)staining.The levels of serum creatinine(SCr)and urea nitrogen(BUN)were measured by automatic biochemical analyzer,and the expression of LC3 and beclin-1 in kidney tissue of rats was detected by Western blot.Results:1.After blunt impact injury,the volume and weight of the kidney increased,the color was dark red,the local subcapsular hematoma,and the renal tissue in the injured area was obviously congestion and edema.2.Hematoxylin-eosin(HE)staining showed that the shape,structure,size,quantity and distribution of glomeruli and renal tubules in normal control group were normal.In the impact injury model group,a large number of red blood cells were found under the renal capsule,red blood cells in glomeruli and renal tubules were significantly increased,renal tubular epithelial cells were edema,granular degeneration,renal interstitial vascular dilatation and congestion,which had a certain relationship with the impact injury of the kidney.In the Bailing Capsule group,there were more red blood cells in renal tissue,edema and granular degeneration of renal tubular epithelial cells,and the degree of renal interstitial vasodilatation was less than that in the impact injury model group.3.Using automatic biochemical analyzer to detect the contents of serum creatinine(SCr)and urea nitrogen(BUN)of rats in each group,the results showed that there were significant differences in the contents of serum creatinine(SCr)and urea nitrogen(BUN)of rats in group A,group B and group C(FSCr=99.034,PSCr=0.000;FBUN=48.262,PBUN=0.000).The contents of SCR and BUN in group B were significantly higher than those in group A(PSCr<0.05;PBUN<0.05),indicating that the renal function of rats was damaged by impact injury,indicating that the model was successfully established.The contents of SCR and BUN in group C were lower than those in group B(PSCr<0.05;PBUN<0.05),but it was significantly higher than that in group A(PSCr<0.05;PBUN<0.05)。4.Western-blot was used to detect the expression levels of autophagy related protein LC3 and Beclin-1 in renal tissue of rats in each group.It was found that the expression levels of LC3 and Beclin-1 in renal tissue of rats in group A,group B and group C were significantly different(FLC3=99.034,PLC3=0.000;FBeclin-1=48.262,PBeclin-1=0.000).The expression of autophagy related protein LC3 and Beclin-1 in the impact injury model group(group B)was higher than that in the normal control group(group A)(PLC3<0.05;PBeclin-1<0.05);The expression of autophagy related protein LC3 and Beclin-1 in group B was lower than that in group C(PLC3<0.05;PBeclin-1<0.05)。Conclusion:1.Blunt impact injury of kidney can lead to the increase of autophagy activity in renal tissue.2.The expression of beclin-1 and beclin-1 in the kidney tissue of rats with blunt renal injury was promoted by the capsule.3.Bailing capsule may play a protective role on the kidney after blunt impact injury by increasing the level of autophagy.
