Effect Of Electroacupuncture Pretreatment Of Neiguan Acupoint At Different Time Points On The Expression Of Autophagy Genes Beclin-1 And LC3 In MIRI Rat

Objective:This experiment is intended to take myocardial ischemia-reperfusion(MIRI)model rats as the object of this study,and to analyze its regulatory effect on MIRI rats by comparing the changes of autophagy gene expression in rats under different time points from electroacupuncture pretreatment intervention at the Neiguan point as the entry point.At the same time,the optimal time point in pretreatment was screened by comparing the cardioprotective effect under different time phases,which laid the theoretical foundation for further clinical application.Methods:1.Forty-eight rats with SD were randomly grouped: divided into blank group,sham-operated group,model group and electro-acupuncture-neiguan group,with 12 rats in each group;the electro-acupuncture-neiguan group was the important observation object,and each group was divided into different subgroups according to different time points,with 3 d and 7 d,with 6 rats in each group.2.The classic rat coronary artery left anterior descending branch perforator ligation was used to create the MIRI model.Hematoxylin-eosin(HE)staining was used to observe the pathological histological changes in rat myocardial tissue;RT-PCR and Western blot were used to detect the expression change levels of autophagy-related expression genes Beclin-1 and LC3 in cardiac myocytes.Result:1.Comparison between groups:(1)3d group: compared with the blank group and sham-operated group,the model group had heavier myocardial injury,disordered myocardial cell order,inflammatory cell infiltration,disrupted mitochondrial structure,and autophagic vesicles;Beclin-1 and LC3-II/LC3-Ⅰ protein expression levels were significantly higher(P<0.01);compared with the model group,the myocardial cell injury in the Neiguan group was less severe,myocardial fibers compared with the model group.Secondly,the protein ratios of Beclin1 and LC3 Ⅱ/LC3Ⅰ were decreased in the endoguan group(P<0.05).(2)7d group: compared with the blank and sham-operated groups,the model group had disorganized myocardial tissue cells with severe mitochondrial vacuolization and relatively blurred structure.The myocardial fibers in the endocannabinoid group were slightly neatly arranged and the damage was reduced.The expression of autophagy-related proteins was significantly higher in the model group(P<0.01);compared with the model group,the expression of autophagy-related proteins appeared to be decreased in the endoguan group(P<0.05).2.Intra-group comparison:(1)Compared with the 3d group,the myocardial damage was less severe and the organelle damage was lower in the endoguan group.(2)Compared with the 3d group,the Beclin1 and LC3-II/LC3-Ⅰ protein expression levels were decreased in the 7d endoguan group(P<0.05).Conclusion:1.When acute myocardial ischemia occurred in rats,electroacupuncture pretreatment of Neiguan acupoint had a protective effect on MIRI model rats by regulating cardiomyocyte autophagy-related genes.2.By comparing the two time points of 3 d and 7 d,the 7 d group was more effective in terms of the difference in myocardial protection during myocardial ischemia-reperfusion.
