Effect Of Bailing Capsule On The Expression Of Nephrin In The Kidney Of The Rats With Diabetic Nephropathy

Objective To investigate the effect of Bailing capsule on the expression of nephrin in the kidney of the rats with diabetic nephropathy.Methods The models of diabetic nephropathy in rats were established by injecting streptozotocin into abdomen,40 male specific pathogen-free (SPF) Wistar rats were randomly divided into 4 groups:normal control group (n=10), diabetic nephropathy group (n=7), Bailing capsule treated group (n=8) and benazepril treated group (n=7).The rats in Bailing capsule treated group were given Bailing capsule (2.5g/kg·d), and Benazepril treated group were given Benazepril tablet(10mg/kg·d) through intragastric administration once daily for 12 weeks. The rats in each group were respectively sacrificed 12 weeks after models established. Blood sugar (BS), kidney weight/body weight(KW/BW),24h urinary protein (24hUpr), Serum creatinine (Scr), Blood urea nitrogen (BUN), and creatinine clearance rate (Ccr) were observed. PAS and Masson staining was used to observe morphological changes of renal tissue. Immunol histochemistry and RT-PCR were used to catch the protein and mRNA expression of Nephrin in renal tissue.Results Compared with normal control group, BS, KW/BW,24hUpr, Ccr, and renal fibrotic lesions deteriorated significantly in diabetic nephropathy group (P<0.05). Treatment with Bailing capsule could improve these parameters except BS. Compared with normal control group, the protein and gene expression of nephrin reduced significantly in the kidney of the rats with diabetic nephropathy (P<0.05). Administration of Bailing capsule significantly restored the protein and gene expression of nephrin, which improved significantly Compared with those in diabetic nephropathy group. Basement membrane thicken, foot process with disorders, foot process fusion, increased mesangial matrix, widened mesangium were observed in model group by light microscope and transmission eletron microscope. Benazepril and Bailing Capsule could improve ultrastructure in kidney of diabetic rats, ameliorate the renal pathomorphology change respectively.Conclusion The kidney injury in the rats with diabetic nephropathy were ameliorated by Bailing capsule to some extent, which may be related to upregulate the protein and gene expression of nephrin.
