| Objective:In this study,we established a model of abdominal impact injury in SD rats to detect blood urea nitrogen(BUN),serum creatinine(SCr)and autophagy-related proteins in renal tissues.Niaoduqing granules were used for intervention to detect the effects of drugs on kidney tissue after injury,and to detect the changes in the expression levels of autophagy related proteins Beclin-1 and LC3,which provided a new direction for the selection of drug treatment after kidney impact injury.Methods:Thirty-five healthy male SD rats were randomly divided into three groups:normal control group(group A,n=5),trauma group(group B,n=15),Niaoduqing Granule group(group C,n=15).The B,C groups were divided into three subgroups of 24 hours,1week and 2 weeks after trauma(n=5),and the kidney impingement injury model was established by BIM-V biological impactor.Group A:anaesthesia only;Group B:After anesthesia impingement,the stomach was gavaged with equal amount of distilled water.Group C:After anesthesia and impingement,5g/kg Niaoduqing suspension was gavaged twice a day.In group A,blood was extracted from the inner canthus vein after anesthesia and the kidney was immediately sacrificed.At corresponding time points after modeling,blood samples from group B and group C were taken from the inner canthus vein to detect creatinine and urea nitrogen,and then the kidneys were sacrificed and taken out for immunohistochemical detection of Beclin-1 and LC3 protein expression,and hematoxylin-eosin(HE)staining was used to observe the pathological changes in renal tissues of rats in each group.Results:1.It can be seen by naked eyes that part of renal parenchyma was damaged in the impingement injury model group,and renal ecchymosis and subcapsular hematoma were formed.It is considered that the model of impingement injury of kidney was established successfully.2.The serum creatinine and urea nitrogen of the blank control group(group A)were increased compared with that of the pure trauma group(group B)and the Niaoduqing group(group C),and the serum creatinine and urea nitrogen of the pure trauma group(group B)were increased more obviously than that of the Niaoduqing group(group C).(PSCr<0.05,FSCr=25.4;PBUN<0.05,FBUN=104.3).The difference was statistically significant.3.Immunohistochemistry showed that Beclin1and LC3 protein expression levels in kidney tissue of A,B and C groups were significantly different(PLC3<0.05,FLC3=111.4,PBeclin-1<0.05,FBeclin-1=131.2),and the protein expression level of the pure trauma group(group B)and the Niaoduqing group(group C)increased gradually with time within 2 weeks(P<0.05),and the increase of protein expression in Niaoduqing group(group C)was more obvious than that in simple trauma group(group B).The difference is significant(P<0.05).4.HE staining showed that the renal capsule was intact in normal control group,and the size,shape,number and distribution of glomeruli and renal tubules were not abnormal.In the trauma group,there was obvious edema and congestion in renal tubules and parenchyma,a large number of inflammatory cells infiltrated in renal interstitium,and medulla hemorrhage.In the Niaoduqing group,the degree of renal tissue injury was slightly less than that in the trauma group,with a small amount of bleeding and inflammatory cell infiltration.Conclusion:1.After kidney impingement injury,the expression levels of autophagy related proteins Beclin-1 and LC3 in kidney tissues increased,and their expression levels gradually increased with time in a certain period of time.2.Niaoduqing granules can increase the expression levels of autophagy related proteins Beclin-1 and LC3 in the damaged kidney to a certain extent,and reduce the levels of serum creatinine and urea nitrogen,and may accelerate the repair of the damaged kidney tissue and the recovery of the damaged renal function by increasing the level of autophagy. |
