Objective: To observe the effect of Xiao Ji Particles of Gastric and Serum pepsinogen on primary Rats with Heat Stagnation.Based on the clinical,starting from basic research,using modern experimental and test methods,Through the different drug groups of stagnation of heat in mice model of gastric acid secretion and serum concentration of pepsin in the original effect,to observe the curative effect of Xiao Ji Particles,From the experimental point of view,we verified the regulation mechanism of the accumulation of particles on the spleen and stomach function of children with stagnation of heat,so as to provide some experimental basis for the further utilization of traditional Chinese Medicine.Methods:Choose 50 healthy Kunming mice,SPF grade,aged 4 weeks,weight(20+2)g,were randomly divided into blank control group,respectively,14rats(group A),model control group 12(B group),Xiaoji granule group 12(group C)and Qingrehuazhi granule group 12 only(group D).Male and female each half,separate adaptive feeding for 3 days,the blank control group(group A)received routine feed,according to physiological saline 0.2ml/10 g orally,2 times a day;the model group(B,C,D group)fedwith special highprotein,highcalorie,homemade concentration of52% milk solution by intragastric administration of 0.2ml/10 g,2 once a day,and intraperitoneal injection of atropine hydrochloride 0.02mg/(0.1mg/ml),2 times a day;All groups were free to eat and drink for 7 days.The general condition,bodyweight,Anal temperature,diet and drinkingwater were observed and recorded every day.Seventh days to create a successful model.The model of mice weight increase speed gradually slow,reducing food intake,increased water intake,Increased rectal temperature,and abdominal distension,restlessness,dry stool and other clinical manifestations,stagnation of heat syndrome in children with similar symptoms,there was significant difference between before and after the statistical model(P<0.05),suggesting that the model is over;A group and B group were given saline gavage of 0.2ml/10g(Appendix 5),C group according to the ?guiding principles of clinical research on new drugs?with Xiaoji granule equivalent dose group0.14g/10 g,group D was given for Qingrehuazhi granule 0.029g/10 g,with a good concentration after intragastric administration by0.2ml/10 g.Mice were given 2 times a day for 1 weeks.After a weekof treatment,all mice can not help but water 12 h,weighing the weight of mice in each group,intraperitoneal injection of pentobarbital sodium anesthesia mice,take blood from the eye,specimens collected by sterile dry tube.3500r/min centrifugal 5min,take the above clean liquid,4 degrees to save for inspection.After killed animal,remove the stomach,gastric juice collected in graduated centrifuge tube,3000r/min centrifugal 15 min,absorb the supernatant of gastric juice analysis.The experiment using enzyme-linked immunosorbent assay(ELISA)determination of pepsin in serum of mice with original concentration;The acidity of gastricjuicewasdeterminedbytitration.Results:(1)The control group showed no abnormal restlessness,weight gain stable,normal intake of food and water,freely,hair bright,normal stool;model group mice at 3 days after modeling,visible irritability,nose scratching action,hair yellow ash,low gloss,easy to get together and curled up body weight of mice,slower growth,eating too less frequently,dry stool,drinking water,after 1 weeks the above symptoms,until all the symptoms eased slightly after intervention;(2)The mice in group D after drug intervention irritability symptoms eased,weight slightly higher than the blank model group of hair growth,improvement,increase food intake,stool soft;mice in group C after drug intervention occasionally catch a nose,irritability,rapid weight gain,eating good water,hair shiny,stool normal.Compared with B group,the weight gain of C group and D group was faster(P<0.05);theweight gain of C group was faster than that of D group(P<0.05).Compared with the B group,C group and D group decreased rectal temperature(P<0.05);compared with the D group C group(P<0.05);rectal temperature decreased compared with B group,C group,D group diet increased(P<0.05);compared with the D group C group diet increased(P<0.05).(3)The content of the original concentration of pepsin in serum: compared with A group B group in serum pepsin concentration decreased(P<0.05);compared with B group,C group,D group of serum pepsinogen in higher concentration(P<0.05);group C serum concentration of pepsin was higher than D group(P<0.05).(4)Comparison of total acid:compared with Agroup,B group of mice decreased gastric acid secretion(P<0.05);compared with B group,C group and D group increased gastric acid secretion(P<0.05);C group of mice gastric acid concentrationwashigherthan D group(P<0.05).Conclusion:(1)During the course of the disease,there was a decrease of gastrointestinal motility in mice;(2)The Xiao Ji Particles has a therapeutic effect on stagnation of heat,and can enhance the gastrointestinal motor function,improve the secretion of gastric acid pepsin content in serum and the amount of the original;(3)The curative effect of Xiao Ji Particles in treating heat stagnation syndrome is betterthan Qingrehuazhi Particles.
