The Effect Of Kechuanning On The NGF By Affect SIRP Alpha In Asthmatic Rat Induced By Respiratory Syncytial Virus

Objective: To study the effect of Kechuanning on the NGF by affect SIRP alpha in Asthmatic Rat Induced by Respiratory syncytial virus,and provides the theoretical basis of never and immue mechanism of Kechuanning.Methods: ?the expression of SIRP alpha and its ligand of CD 47 in asthma and the intervention effect of Kechuanning: From 30 Wistar rats randomly selected 5 for normal group,in addition to the normal groups,intransal administrationi other groups with 50 ul RSV,use Ovalbumin to made rats sensitization 2 weeks.Randomly divided the 25 rats into 5 groups,each group of five,ratsrespectively,asthma rats group(model group),SIRP alpha antibody intervention group(SIRP alpha antibody group),different doses of kechuanning group(kechuanning group of ??).,after the completion of model building,normal rats group will ? not be any processing,model group with saline lavage;intraperitoneal injection of anti-SIRP? group with anti-SIRP?.Give a gavage of fifferent doses of kechuannig to kechuanning groupsfor 2 weeks.Use PCR method to detect the expression of SIRP? in spinal ganglion and spinal dorsal horn from C7 to T5 of model ras.The effects of SIRP alpha and Kechuanning to NGF :From 60 Wistar rats randomly selected 6 for normal group,in addition to the normal groups,intransal administrationi other groups building methods like above.Randomly divided the 54 rats into 9 groups,each group of six,espectively,the asthma rats group(model group),SIRP alpha antibody intervention group(SIRP alpha antibody group),SIRP alpha intervention group(SIRP alpha group),different doses of kechuanning group(kechuanning group of ? ??),SIRP alpha + different doses of kechuanning intervention group(combination group of).after the completion of model building,? ? ?normal rats group will not be any processing,model group with saline lavage;intraperitoneal injection of anti-SIRP? group with anti-SIRP?.SIRP alpha intervention group(SIRP alpha group),Give a gavage of fifferent doses of kechuannig to kechuanning groups,intraperitoneal injection of alpha SIRP alpha team with SIRP alpha for 2 weeks.Use PCR method to detect the expression of NGF in spinal ganglion and spinal dorsal horn from C7 to T5 of model ras.Results :(1)SIRP alpha in normal group content is the lowest,Kechuanning grioup of ? is more than normal group,but there was no statistically significant difference between the two groups(p > 0.05),and compared normal group with the rest groups,the difference was statistically significant(p < 0.05);Compared with model group,the SIRP alpha in Kechuanning groups and antibody group were less(P < 0.05);Compared with the antibody group,the SIRP alpha in Kechuanning groupof ?,? there was no statistically significant difference(P > 0.05);compared with antibody group the Kechuanning ? group has lower contents of SIRP alpha antibody(P < 0.05).Compared Kechuanning between each group,the SIRP alpha from most to lest is Kechuanning ? group < Kechuanning group of ?to Kechuannig group of ?,the difference was statistically significant(P < 0.05).The CD47 in normal group is the lowest,but compared with Kechuanning group of ?,SIRP alpha antibody group the differences had no statistical significance(P > 0.05);The remaining groups compared with the blank group,CD47 were increased(P < 0.05);Compared with model group,CD47 in groups of Kechuanning and antibody were less(P < 0.05);The kechuanning groups compared with antibody group,CD47 in Kechuannig group of ? is the lowest,but compared with antibody group,there was no statistically significant difference(P > 0.05).Kechuanning group of ?,? content more CD47 than antibody group(p < 0.05);There was no statistically significant difference between kechuannig group I and III(P > 0.05).(2)NGF in Normal group is least,but there was no statistically significant difference when compared with the Kechuanning group of ?(p > 0.05);Compared with model group,the antibody group and kechuanning groups have less NGF in have different degrees.(p < 0.05);compared Antibody group to Kechuanning groups,Kechuanning group of ? has least NGF(P < 0.01),Among Kechuaning group of I ?III and the antibody group,there was no statistically significant difference.Joint group ? content least NGF in all joint groups(P < 0.05),But higher than Kechuanning group of ?(P < 0.05).Compared joint group of I with joint group III,there was no statistically significant difference(P > 0.05).Conclusion :(1)SIRP alpha and its ligand CD 47 participate in the onset of asthma,and Kechuaningcontrol has effect on it.(2)Kechuanning and SIRP alpha antibody can influence the expression of NGF plays,and the efforts of Kechuanning can be weaked by SIRP alpha antigen,that means Kechuanning could through regulating SIRP alpha and influence the expression of NGF plays.(3)The effect of Kechuanning on the NGF by affect SIRP alpha in Asthmatic Rat Induced by Respiratory syncytial virus maybe the way to improve for neurogenic inflammation.
