Kechuanning Oral Liquid Regulates The Effect Of Sema4D On NGF In RSV Combined OVA Asthma Model Rats

Objective:Observe the expression of Sema4 D and NGF in lung and visceral sensory afferent nerves of asthmatic rats,and explore the relationship between Sema4 D and NGF and the influence of Kechuanning Sema4 D on NGF,so as to provide basis for further elucidating the mechanism of the overall regulation of Kechuanning.Method:Effect of Sema4 D on NGF Expression in the Onset of Asthma and the Intervention Effect of Kechuanning: 72 Wistar Rats of Young Age,12 were selected as normal group according to the random number table method,the other groups were used as model group.The model group was modeled according to the RSV compound OVA asthma model of young rats.After 14 days of sensitization,6 rats were randomly selected from the normal group and the model group to observe whether the mental state,heart rate,respiration,and airway hyperresponsiveness were determined.After the asthma model was completed,the rats after successful modeling were divided into9 groups according to the random number table method,with 6 rats in each group,namely,the asthmatic rat group(model group)and the asthmatic rat Sema4 D intervention group(Sema4D group).),Asthma rat Sema4 D antibody intervention group(Sema4D antibody group),asthmatic rat low-,middle-and high-dose Kechuanning intervention group(kechuanning I,II,III group),asthma rat Sema4D+different doses of Kechuanning Intervention group(group I,II,III).In addition to the normal group of 6,only the other 9 groups of rats were given drug intervention according to group conditions: Sema4 D group was given Sema4 D intraperitoneal injection,Sema4 D antibody group was given Sema4 D antibody intraperitoneal injection,Kechuanning I,II,III group were given Kechuanning Low,medium,and high doses of gavage,Sema4 D plus different doses of Kechuanning intervention group were given Sema4 D intraperitoneal injection and Kechuanning low,middle,and high dose gavage(according to the clinical dose-response observations to determine the high dose of Kechuanning oral solution 1ml/100 g,middle dose0.3ml/100 g,low dose 0.033ml/100g)once daily for 2 weeks..After two consecutive weeks of treatment,lung tissue and spinal cord tissue were removed from rat models.Real-time PCR,immunohistochemistry and Western blotting techniques were used to detect the expression changes of NGF.Result:Effect of Sema4 D on the Expression of NGF in the Onset of Asthma and the Intervention Effect of Kechuanning1.Effect of Sema4 D on NGF in the pathogenesis of asthma: Compared with the model group,NGF expression in Sema4 D group was significantly higher(P<0.01),and the difference was statistically significant.Compared with the model group,the expression of NGF in the Sema4 D antibody group was significantly reduced,p<0.01,and the difference was statistically significant.2.The influence of Kechuanning on the NGF in asthmatic patients: 1 The expression of NGF in the blank group was the least.The expression of NGF was increased in each group compared with the blank group,P<0.01,and the difference was statistically significant.2Compared with the model group,the NGF levels in the Sema4 D antibody group and the Kechuanning group all decreased,p<0.01,and the difference was statistically significant.Compared with the model group,NGF expression was slightly higher in the combination I group,p>0.05,and the difference was not statistically significant.Compared with the model group,the expression of NGF in the combination II,III groups was reduced,P <0.01,the difference was statistically significant.Compared with the Kechuanning group,the expression of NGF in the 3Sema4 D antibody group was lower in the Kechuanning II and III groups,P<0.01,and the difference was statistically significant.The expression of NGF in Kechuanning I group was higher than that in the antibody group,P<0.01,and the difference was statistically significant.4Compared with Kechuanning III group,NGF expression in model group,Sema4 D group,Sema4 D antibody group,Kechuanning I group,and combination I,II,III group were significantly increased,P<0.01,the difference was statistically significant.significance.Compared with Kechuanning III group,the NGF content in Kechuanning II group was slightly lower(p>0.05),and the difference was not statistically significant.Conclusion:1.In the onset of asthma,Sema4 D promotes the expression of NGF and aggravates the development of asthma.2.When asthma attacks,Kechuanning oral solution can inhibit the expression of NGF,Sema4D's promoting effect on NGF can be weakened by Kechuanning,and Kechuanning's middle and high dose group has the best regulation effect,namely Kechuanning oral.The fluid can affect the expression of NGF by regulating Sema4 D.3.The effect of Kechuanning Oral Liquid on the NGF of rats induced by RSVinduced by Sema4 D can provide a basis for further elucidating the mechanism of the overall regulation of Kechuanning.
