Objective:Observation to the effect regulated by Kechuanning Oral Liquid of Sema4 D on the function of HPA axis in the asthma model rats induced by virus on animal experiment method,and to probe the mechanisms of neuroendocrine immune network of Kechuanning oral liquid in asthma,and further expounds the overall regulation of traditional Chinese medicine using experimental data.Methods : Randomly selected from 72 young Wistar rats,and according to random number table method to select 12 as normal group,the rest of the group as a model group,model group,in accordance with the young rats RSV composite OVA asthma model building,the sensitization after 14 days,from the normal group and model group rats were randomly selected from 6 only observe mental state and the heart rate,breathing and detection of airway hyperresponsiveness judgment model was successful.After the model of the asthma model was completed,the rats who succeeded in the model were divided into 9 groups according to the random number table method,6 of each group.asthma rats group(model group),Sema4 D interfered with the asthma rats group(Sema4D group),Sema4 D antibody interfered with the asthma rats group(Sema4D antibody group),Low,medium,high doses of Kechuanning oral liquid interfered with the asthma rats group(Kechuanning oral liquid?????group),Sema4D+different doses of Kechuanning oral liquid interfered with the asthma rats group(united????? group).In addition to the normal group,the other 9 groups were given drug intervention by grouping: Sema4 D group was given Sema4 D intraperitoneal injection,Sema4 D antibodies group was given Sema4 D antibodies intraperitoneal injection,Kechuanning oral liquid?????group was given Kechuanning oral liquid low,medium and high dose lavage,the united?????group was given Sema4 D intraperitoneal injection and Kechuanning oral liquid low,medium and high dose lavage(according to the relationship between the clinical effects of observation Kechuanning oral liquid of high dose of 1 ml / 100 g,the dose of 0.3 ml / 100 g,low doses of 0.033 ml / 100 g).1 times a day,a total of 14 days.After treatment,immunohistochemical method was used to detect c-fos protein in rats and the contents of SP in PVN in rats and ACTH and CORT in ME were detected by radioimmunoassay.Results :(1)The influence of Sema4 D on the HPA axis during the onset of asthma:(1)Compared with normal group,Sema4 D group in c-fos protein expression,higher content of SP in PVN,Sema4 D group CRH content,content of ACTH,CORT content decreased significantly,there are significant difference statistically significant(P < 0.01).(2)Compared with model group,the SP content of Sema4 D group was obviously higher,there are significant difference statistically significant(P < 0.01),CRH content decreased,there was no statistically significant difference(P > 0.05),the ACTH content of in peripheral blood,CORT content decreased obviously,there are significant difference statistically significant(P < 0.01),and Sema4 D antibody group c-fos protein expression is a bit higher,CORT content is slightly lower,there was no statistically significant difference(P > 0.05);SP content was significantly decreased,CRH content and ACTH content were significantly increased,and the difference was statistically significant(P < 0.01).(2)The effects of Kechuanning oral liquid on HPA axis by regulating Sema4 D during asthma onset:(1)Compared with normal group,each group of rats in c fos protein expression,higher content of SP in PVN,in addition to Kechuanning oral liquid ? group c-fos protein expression,the remaining differences have a significant statistical significance(P < 0.01);Kechuanning oral liquid?and united ? group content of ACTH,CORT content decreased significantly,the model group CRH content,ACTH content decreased significantly,content of ACTH united ?,?group were significantly lower,Sema4 D antibody group,Kechuanning oral liquid ?CORT content decreased obviously,there are significant difference statistically significant(P < 0.01),Kechuanning oral liquid ? group ACTH levels drop,the difference was statistically significant(P < 0.05).(2)Compared with model group,normal group,Kechuanning oral liquid ? group,the united ? group c-fos protein expression,SP content significantly decreased,there are significant difference statistically significant(P < 0.01),Kechuanning oral liquid ? group c-fos protein expression decreased,the difference was statistically significant(P < 0.05),Normal group,Sema4 D antibody,Kechuanning oral liquid ?,?,? group,the united ?group CRH content,ACTH content and the ACTH content of united ? group was obviously higher,there are significant difference statistically significant(P < 0.01),united ? group CRH content increases,the difference was statistically significant(P< 0.05),but the ACTH content of united ? group is slightly higher and more groups;CORT content lifting is differ,there was no statistically significant difference(P >0.05);(3)Compared with the Sema4 D group,c-fos protein expression and SP content of the other groups were significantly reduced,and the difference was statistically significant(P < 0.01).Between groups of CRH content,ACTH content,CORT content levels have increased,in addition to the model group of CRH content and the concentration of Sema4 D antibody group,Kechuanning oral liquid ?,CORT content outside,the rest all have significant statistical significance(P < 0.01);(4)Compared to Kechuanning oral liquid ? group,Kechuanning oral liquid ? group c-fos protein expression,united?SP content increases,the difference was statistically significant(P < 0.05),model group,Kechuanning oral liquid?group c-fos protein expression,content of SP were significantly elevated,Sema4 D antibodies,untied?,?group c-fos protein expression were increased significantly,content of SP normal group was obviously lower,with significant difference statistically significant(P < 0.01),Kechuanning oral liquid ? group CRH content,ACTH content,CORT content,Model group CRH content,content of ACTH and united ?,?,? group content of ACTH decreased obviously,there are significant difference statistically significant(P < 0.01),united ? group CRH content,CORT content and Sema4 D antibody group;CORT were decreased,the difference was statistically significant(P < 0.05).Conclusion : 1.The function of HPA axis in rats was inhibited when asthma occurred,and the inhibition of HPA axis could be improved by Kechuanning oral liquid.2.Sema4 D is involved in the pathogenesis of asthma,which can further inhibit the release of HPA axis endocrine hormones by stimulating the expression of c-fos protein in rats' brain and release of SP,thereby aggravating asthma.3.Kechuanning oral liquid can adjust the function of HPA axis of Sema4 D of asthma model rats,which the effect is better,Kechuanning oral liquid medium and high dose group.It is suggested that Kechuanning oral liquid can plays a preventive role in asthma by improving the inhibition of Sema4 D on the HPA axis of asthmatic rats.
