The Effect Of Kechuanning Oral Liquid On The Expression Of P13k In The Rat Model Of Asthma Induced By Virus

Purpose:research that Kechuanning Oral liquid impact on the expression of P13k in the cell of CD4+T、CD8+T about RSV-induced asthma, to set forth the mechanism about this medicine preventing and treating this kind of asthma, and to provide experimental evidence for finding new way to help asthma better.Methods:make40rates into4groups randomly by weight, these are PBS control group, and others are model groups, which are PBS group, dexamethasone with salbutamol group, and Kechuanning Oral liquid group. Give model groups1ml sterile antigen solution injection in three different parts (0.25ml in medial side of the both lower limbs and0.5ml in the abdominal cavity), which contain1mg OVA and100mg amphojel in the first and third day for allergization, and then in the21th、35th、49th day drip the RSV50ul (its titre1.0×106) in nasal cavity.PBS control group was allergized and boosted by PBS.Separate the monocyte in peripheral blood in the four groups by routine and CD4+T、CD8+T cells by bead combination method. Then add the cell suspension of model groups into three culture plates, mark them group A, group B, and group C respectively, at the same time add PBS10ul in the PBS control group. By means of immunoflurescence method, we can assess the expressive level of PI3K-110α in T cell by flow cytometry according to the amount of the fluorescence which is boosted by the marked-PE antibody.Results:1) the successive build of model there appears obvious anthropoid acute asthma seizure in-asthma groups.HE stain of lung pathological section show:there are no thickening areflue epithelial cells and no perivascular and airflue inflammation; and there are obvious inflammatory cell infiltration especially neutrophilic granulocyte in areflue and around the blood vessels, partly bronchus epithelial cells drop off incompletely.2) Flow cytometry results show:(1) CD4+T. CD8+T positive rate comparision in each groups:The positive rate is higher apparently in model groups than others groups, and the differences have statistical significance (allP<0.01).(2)The comparison of mean fluorescence intensity of P13K on the cells of CD4+T\CD8+T in every groups:the intensity is higher in model one than others, and the differences have statistical significance (all P<0.01);(3)The positive rate of CD4+T、CD8+T has no correlation with the mean fluorescence intensity of P13K on the cells of CD4+T、CD8+T by pearson’s correlation analysis.Conclusion:1)CD4+T、CD8+T cells involves in the asthma immunity balanced modulation, and the expression of P13K on the cells has something in inducing asthma; dexamethasone with salbutamol and Kechuanning Oral liquid can arrest the expression of P13K on the CD4+T、 CD8+T cells, which may be one of the immunologic mechanism of treating asthma.
