Purpose:This subject is mainly about the preparation technology,quality standard and pharmacodynamics of Ke chuan ning oral solution,which provides a theoretical basis for the industrial production and clinical application of Ke chuan ning oral solution.Method:1.Study on Preparation Process of Ke chuan ning oral solution.The extraction amount of baicalin in scutellaria,ephedrine and pseudoephedrine in ephedra,was used as the inspection index,L9?34?orthogonal test was adopted,and extraction solvent,extraction time and extraction times were taken as the investigation factors to optimize the extraction process of Ke chuan ning oral solution.Optimize the process of Ke chuan ning oral solution preparation to get the best extraction scheme.2.Study on the quality standard of Ke chuan ning oral solutionTLC was used for identification studies;HPLC was used for content determination studies;Ke chuan ning oral traits and examinations;and a draft standard for Ke chuan ning oral solution was established.3.Pharmacodynamic study of Ke chuan ning oral solution?1?Ammonia coughing methodAmmonia coughing method is commonly used experimental method for antitussive experiment.Concentrated ammonia has a strong volatility and irritation.Concentrated ammonia was sprayed into the closed bell jar.After the mouse inhaled concentrated ammonia,the respiratory receptors were stimulated,and the reflected ground could cause coughing.Antitussive drugs can suppress the cough center or reduce the sensitivity of respiratory receptors,and thus play a role in antitussive.The effect of antitussives is reflected in the incubation period of the cough and the number of coughs in a certain period of time.Therefore,in this experiment,ammonia was used to induce cough to establish an animal model.Cough latency and the number of coughs in a certain period of time were used as indicators of antitussive effect.?2?Mouse phenol red excretion methodThe phenol red excretion method is one of the commonly used experimental methods in the expectorant experiment.After intraperitoneal injection of phenol red mice,the excretion of phenol red in the airways of mice can be measured from the characteristics of airway excretion to determine the effect of Kechuanning oral solution on airway permeability and secretion volume.Therefore,the excretion of phenol red was used as an observation index to reflect the phlegm-effect of Ke chuan ning oral solution.?3?Guinea pig acetylcholine chloride-histamine histidine induced asthma methodThe asthma medication is acetylcholine chloride-phosphate histamine.The asthma medication was sprayed into the closed glass bell jar through the spray device,causing the guinea pig to suffocate and suffocate,resulting in twitch and fall.The establishment of this animal model can be used to observe the bronchial smooth muscle relaxation of drugs.Therefore,the the latent period of the guinea pig's twitching as observation that Baicalin injection volume has good linearity in the range of 0.086125 to 1.378?g,r=0.99997.The precision,repeatability and stability of the content determination method were all good.The recovery rate was 98.11%,RSD.=1.92%;Ephedrine hydrochloride injection volume has good linearity in the range of0.10561.32?g,r=0.99998.The precision,repeatability and stability of the method were all good,the recovery was 101.291%,RSD=0.306%.Pseudoephedrine hydrochloride injection volume had a good linearity in the range of 0.11121.39?g,r=0.99999.The precision,repeatability and stability of the content determination method were all good.The recovery rate was 99.760%and the RSD was 2.19%.3.Pharmacodynamic experiments showed that,after randomization,the high-dose and low-dose groups of Ke chuan ning oral solution could prolong the incubation period of cough in ammonia-induced cough and reduce the number of coughing in a certain period of time.Therefore,Ke chuan ning oral solution has antitussive effect.The excretion amount of phenol red in the high-dose Ke chuan ning oral solution group was greater than that in the blank control group,indicating that the amount of phenol red in the respiratory mucosa was increased under the influence of Ke chuan ning oral solution.Therefore,Ke chuan ning oral liquid can affect the airway permeability and the amount of exudate,that is,Ke chuan ning oral solution has a phlegm effect.The fall time of the animals in the high-dose Ke chuan ning oral solution group was longer than that of the blank control group,indicating that Ke chuan ning oral solution had antiasthmatic effects.Conclusion:1.The preferred prosess of Ke chuan ning oral solution is stable and reliable,which is suitable for industrial production requirements.2.Established the TLC identification and content determination method of Ke chuan ning oral solution,formulated the quality standard of Ke chuan ning oral solution,and the traits and examination of Ke chuan ning oral solution,the draft of Ke chuan ning oral solution quality standard.3.Through the experimental study,Ke chuan ning oral solution has antitussive,expectorant and antiasthmatic effects and provides basis for Ke chuan ning oral solution for cough and asthma.
