Objective:Observation to Kechuanning intervention SIRP alpha asthma model rats Th1 / Th2,the influence of asthma in the nervous system and the interaction mechanism of the immune system,further clarify neurogenic inflammation mechanism of asthma and Kechuanning regulating mechanism.Methods:(1)The asthma onset SIRP alpha and its ligand expression of CD 47 and Kechuanning oral liquid on its expression.30 young Wistar rats,and randomly selected 5 for the normal group,the rest of the animals in the young rats RSV composite OVA asthma model building,the sensitization of 2 weeks later,the rats by observing the spirit and heart rate,breathing to test model was successful.Die is to be built upon the building,the success of the animals are divided into 5 groups according to random number table method,each group 5,only the asthma rats group(model group),asthma rats SIRP alpha antibody intervention group(SIRP alpha antibody group),asthma rats to Kechuanning the rather low dose treatment group(group to Kechuanning?),asthma rats to Kechuanning the dose in the treatment group(group to Kechuanning?),asthma rats to Kechuanning the rather high dose treatment group(group to Kechuanning?).In addition to the normal group,the rest of the five groups of rats respectively to medicine(SIRP alpha antibody group shall be SIRP alpha intraperitoneal injection of antibodies and other groups to give,to Kechuanning?,?,? ? ?group),asthma rats SIRP alpha + different doses to Kechuanning intervention group(joint ? ? ?group),the 6 groups have to Kechuanning lavage treatment,according to the relationship between the clinical effects of observation to Kechuanning of high dose of 1 ml / 100 g,the dose of 0.3 ml / 100 g,low doses of 0.033 ml / 100 g,1 times a day,for 2 weeks,until death.Asthma rats SIRP alpha intervention group(SIRP alpha group),asthma rats SIRP alpha + different doses to Kechuanning intervention group(joint ? ? ?group),the four groups are given by intraperitoneal injection of SIRP alpha;Asthma rats SIRP alpha antibody intervention group(SIRP alpha antibody group)to give SIRP alpha intraperitoneal injection of antibodies.Intraperitoneal injection of 1 times a day,a total of 2 weeks.Detection in the animal model of rats serum IFN-?,IL-4 expression changes: inflammation factors such as by using enzyme-linked immunosorbent(ELISA)method.Result:1.The asthma onset SIRP alpha and ligand expression of CD47:(1)Compared with normal group,the group to Kechuanning ?SIRP alpha and its ligand expression of CD47 slightly increased,but there was no statistically significant difference(P > 0.05);(2)Compared with normal group,Kechuanning group,model ?group,the antibody group,Kechuanning the ?group,Kechuanning ?SIRP CD47 alpha and ligand expression were significantly higher,the difference was statistically significant(P < 0.05);(3)Compared to Kechuanning group,Kechuanning group,? ?Kechuanning ?SIRP CD47 alpha and ligand expression were significantly higher,the difference was statistically significant(P < 0.05);(4)Compared with SIRP alpha antibody group,Kechuanning ?group SIRP alpha on mRNA expression increased slightly,to Kechuanning ?group SIRP alpha and CD47 protein expression level slightly lower,but there was no statistically significant difference(P > 0.05),Kechuanning ?SIRP alpha group,model group and the expression of ligand CD47 were significantly increased,and the difference has significant statistical significance(P < 0.01);(5)Compared with model group,normal group,the SIRP alpha antibody group,Kechuanning ?group,Kechuanning ?group,Kechuanning ?SIRP alpha and ligand CD47 expression were significantly lower,the difference was statistically significant(P < 0.05).Among them to Kechuanning group SIRP alpha and CD47 at ?the level of protein expression was slightly lower than SIRP alpha antibody group,but there was no statistically significant difference(P > 0.05).2.The asthma onset SIRP alpha of Th1 / Th2 immune regulation(IL-4 and the variation in content of IFN-gamma)and the intervention effect to Kechuanning oral liquid.(1)Normal group,the antibody group,Kechuanning ?group were slightly different,but there was no statistically significant difference(P > 0.05);(2)And normal group,Kechuanning ?group,antibodies,antigens,combined ?,?,?group,Kechuanning ?,IL-4 ?group were significantly increased,IFN-? were significantly lower,the difference was statistically significant(P < 0.05);(3)Compared with model group,the joint group IL ?-4 content is slightly lower,IFN-? is a bit higher,but there was no statistically significant difference(P > 0.05)joint ?,higher contents of IL-4 ?and IFN-significantly lower gamma,antibody group,Kechuanning ?IL-4 content decreased obviously and IFN-? content increased significantly,the difference was statistically significant(P < 0.05);To Kechuanning ?,IL-4 group was obviously higher but IFN ?-? content is slightly lower,there was no statistically significant difference(P > 0.05);The content of IL-4;(4)Antigen group was obviously higher than that of other groups,but IFN-? obviously lower than other groups,and the difference was statistically significant(P < 0.05);(5)Compared to Kechuanning group,IL ?-4 levels to Kechuanning group ?was obviously lower and higher IFN-?,and the difference was statistically significant,joint ?,higher contents of IL-4 and IFN ?-? is reduced,the difference was statistically significant(P < 0.05);IL-4 to Kechuanning ?group content is slightly lower,IFN-? is a bit higher,but there was no statistically significant difference(P > 0.05);Contents of IL-4 joint ?slightly elevated,IFN-? slightly lower,but there was no statistically significant difference(P > 0.05).Conclusion:(1)The asthma onset SIRP alpha and ligand CD47 expression significantly increased,to Kechuanning oral liquid can inhibit SIRP alpha and the expression of ligand CD47,dose group effect is best..(2)Asthma onset of Th1 / Th2 imbalance obviously,Kechuanning oral liquid could through intervention to improve SIRP alpha Th1 / Th2 imbalance,thus improve asthma airway inflammation.(3)To Kechuanning oral liquid can effectively improve asthma Th1 / Th2 imbalance,including dose treatment group effect is best.
