In recent years, Surface Plasmon Resonance (SPR) technique, characterized by high sensitivity, dispensing with sample pretreatment, less sample volume requirement and fast response, has been gotten more and more attention of experts and scholars, and it has been widely applied in biochemical application, food safety administration and medical detection. As the increasing requirement of biological and medical application, the miniaturization multi-channel and automation of SPR systems are the current trends of SPR sensor technology. In this thesis, the main task is designing an angle-modulated SPR sensor and analyzing the sensitivity of the instrument.We present the superiorities, application, history and current situation of SPR technology. We get the conditions of exciting SPR phenomenon by SPR’s theoretical analysis. According to the modeling and simulation of the Surface Plasmon Resonance system based on Kretchmann prism structure, factors such as source line width, optical dispersion of prism, slide glass’s thickness, the thickness of metal film affecting SPR curve has been analyzed. Then, an angle-modulated SPR sensor designed by ourselves has been presented with the aspects such as mechanical circuit design, software program. The mechanical circuit design involves the coordination between linear CCD and turntable, drive circuit of turntable, optical design of He-Ne laser and prism, the sampling system architecture. The software program based on Lab VIEW platform includes controlling the stages and data acquisition, data processing. At last, we can test the characteristics of the samples.With an angular resolution is set to0.03°, the experimental results on the resonance angle monitor of saline solution with different refractive index show that the relationship between the refractive index and SPR resonance angle is well described by linear fitting, and system detection sensitivity is up to1.767×10-4RIU. With optimizing system angular resolution more, it is clear that system detection sensitivity can be further improved.This thesis made completed some theoretical and experimental investigations about SPR instrumentation, which provides solid foundation for future SPR sensor improvement in our group.
