An optical-fiber sensor based on surface plasmon resonance (SPR) effect is a novel sensor which surface plasmons are optically excited base on optical fiber core. This design possesses not only the characteristic of high sensitivity of SPR, but also many advantages of optical fiber, e.g,small size and suitable for remote telemetering,it may be a useful sensor device in many areas, especially in gene mutation detection, biological determination of molecular reaction dynamics, the research of structure and activity, nucleic acid research and industrial wastewater emission control and so on.Firstly, the configuration design of optical fiber SPR sensor system and principle of light Intensity–modulation are discussed in detail. The factors which affect light reflectivities are analyzed, such as the fibers their diameters, length, metallic film thickness. The analytical results provided the theory foundation for making the optical fiber SPR sensor system based on reflect light intensity modulation.Secondly, Through making the optical fiber SPR sensor system based on reflect light intensity modulation, singles from electrical measurement are acquired based on visible interface of Labview. Using it, the relationship between refractive index and light intensity are analyzed by measuring different refractive index of liquid mediums. Thereby, provided support to instrument miniaturization.Finally, Resonant wavelength changes when refractive index changes between metal and solution. According to this theory, a novel optic-fiber biosensor based on surface plasmon resonance (SPR) is made and developed. Thiolated oligonucleotide is immobilized on the surface of gold-film probe with self-assembly. A novel optical fiber SPR nucleic acid biosensor which can recognize special pilot sequence is developed. And it is also used to hybridization experiment of oligonucleotide probe.
