In the past decades, Surface Plasmon Resonance (SPR) technology is a very important analysis tool in many fields such as biomedical analysis, food safety inspection and chemical monitoring. Fiber-optic SPR sensor has attracted more and more interests because of it’s a series of advantages of smaller sample volume requirement, non-markable analysis of biological sample, small volume, multiple parameters sensing, easy integration and remote sensing capabilities. In this thesis, we present our investigation works on the development of a noval optic-fiber SPR sensing system and its application in specific recognition of protein molecules, the major works are listed as follows:Firstly, parameters affecting fiber-optic SPR sensor performance are analyzed by calculation and simulation, to optimize the sensing system design, based on the theoretical foundation of fiber-optic SPR model; SPR wavelengths of the designed sensor for refractive index1.33-1.43are during the range of visible wavelengths; and while sensitivity is10-5RIU/nm. Secondly, according to the above analysis results, single-channel SPR sensor is manufactured and at the same time, we completed the development of a fiber-optic SPR sensing system including light source, a home-made Y-shaped fiber-optic coupler, spectrometer, peristaltic pump, etc. Secondly, in order to improve the signal demodulation’s precision, data analysis system based on the software Labview, which is the software development platform of virtual instrument, is developed; and it can complete a series of operations containing the real-time data acquisition, data analysis, graphical display and data storage; experiments about prime SPR sensing system and fiber-optic detection system are under investigation, respectively; results prove the feasibility of the analysis system software; all the experiment research include refractive index sensing detections of sucrose solutions and PBS buffer solutions and specific recognition of protein molecules; experiment results show that fiber-optic SPR sensor has smart response spectrum. the relationship between the refractive index and SPR wavelength is well described by linear fitting within the measurable refractive index range, and the sensor can be successfully applied in biochemical experiments, especially the detecting molecular dynamics process. Finally, we present a novel dual-channel fiber-optic SPR sensor with multi-alternating metal layers and silver and gold film of different thickness for biochemical analysis, to improve the sensing performance and realize multi-parameter measurement. Based on the fundamental SPR theory of fiber optic sensing technology, the sensor is successfully put in the lab-scale experiments for dual-parameters refractive index and temperature measurement.This thesis demonstrates our theoretical and experimental investigations of fiber-optic SPR sensor system and its application in specific recognition of protein molecules, which provides foundation for future SPR technology improvement and practicability in our group.
