Purpose:To discuss traditional Chinese medicine awareness of type 2 diabetes mellitus complicated with cognitive impairment.Through experimental research, to observe TangZhiQing Granules effect on blood glucose, blood lipid levels, ability of learning and memory, and IRS-1 & IRS-2 expression in the hippocampus in type 2 diabetic model rats and exploring its mechanism.Methods:Use high sugar and high fat diet combined with low-dose Streptozotocin to establish rat model of type 2 diabetes.Type 2 diabetic model rats were randomly assigned to model groups, pioglitazone group, TZQ high-dose group, TZQ middle-dose group. In addition set a normal group. After 4 weeks of medication, observed in rats following indicators:① general state;②glucose;③lipids;④IRS-1 and IRS-2 expression in hippocampus of rats; ⑤ Morris water maze test.Results:①Blood glucose:After treatment, TZQ middle-dose group, pioglitazone group fasting glucose compared with before treatment were statistically significant (P<0.05), were statistically compared with model group (P<0.05).After treatment, TZQ high-dose group fasting glucose compared with before treatment had significant difference (P<0.01), there are significant differences compared with model group (P<0.01).②Lipids:After treatment, TZQ high-dose group, TZQ middle-dose group, pioglitazone group TC, TG, LDL-C level lower than model group, a statistically significant (P<0.05), after treatment, TZQ high-dose group, TZQ middle-dose group, pioglitazone group HDL-C level compared with model group had no statistically significant (P>0.05).③ Water maze test:Each set of escape latency trends over time; but the time factor does not differ depending on the Group (P>0.05). Experimental results of space exploration, after dropping the platform,the number of Pioglitazone group and TZQ middle-dose group across platform is more than model group, there was a statistically significant (P<0.05),the number of normal group and TZQ high-dose group across platform is more than model group, there was a significant difference (P<0.01).Normal group and TZQ high-dose group cross platform-quadrant swimming time/total time higher than the model group, a statistically significant (P<0.05).④Immunohistochemical staining:Model group IRS-1 levels in rat hippocampus lower than the normal group, and compared the two groups was statistically significant (P<0.05), Pioglitazone group, TZQ high-dose group IRS-1 levels in rat hippocampus higher than model group, there are significant differences (P<0.01), TZQ middle-dose group IRS-1 levels in rat hippocampus higher than the model group, a statistically significant (P<0.05). Model group, pioglitazone group, TZQ high-dose group, TZQ middle-dose group IRS-2 levels in rat hippocampus higher than normal group, but not statistically significant (P>0.05), and there was no statistically significant between each group (P>0.05).Conclusion:TangZhiQing Granules can significantly reduce blood sugar, blood lipid levels of the high sugar, high fat diet and low-dose Streptozotocin-induced model rats of type 2 diabetes. TangZhiQing Granules has the effect of promoting the learning and memory ability of rats. The mechanism may be related to improve the metabolism of glucose and lipid, improve the expression of IRS-1 in hippocampus, repaire brain insulin signaling transduction pathways.
