Experimental Research Of TangZhiQing Granules Effect On Myocardial CTGFmRNA And TGF-β1mRNA Expression Of Type 2 Diabetes Mellitus Rats

Objective:To discuss traditional Chinese medicine awareness of type 2 diabetes mellitus complicated with diabetic cardiomyopathy.Through observing the bloodcose、blood fat、the expression of CTGFmRNA and TGF-β1mRNA in the myocardium of type 2 diabetes mellitus rats induced by STZ,it’s aim to investigate the mechanism of TangZhiQing Granules preventing and curing Diabetic cardiomyopathy.so it can apply the experiment statistics for clinic.Methods:Take 50 SD rats,10 for normal group and 40 for experimental groups by random.The experimental groups were fed by high sugar and high fat diet for about 8 weeks.Later they were injected with low-dose Streptozotocin(30mg/kg)to establish rat model of type 2 diabetes trough peritoneal cavity.Then group the Type 2 diabetic model rats into four groups,including model group,irbesartan group,TZQ high-dose group and TZQ middle-dose group.With interference for about 4 weeks,the experiment ended.Last observed the general status,fasting blood glucose,blood fat and the espression of CTGFmRNA and TGF-β1mRNA in the myocardium with the method of RT-PCR.Results:①Compared TZQ high-dose group,TZQ middle-dose group with model group after treatment,the general conditions of rats were ameliorated.And descent of body weight was reved down(P<0.05).②Compared the blood glucose level of TZQ high-dose group,TZQ middle-dose group after treatment with prior treatment,there was statistically significant.And when compared with model group,there was statistically significant(P<0.05).And the high-dose group was more obvious(P<0.01).③The TC,TG and LDL-C level of TZQ high-dose group,TZQ middle-dose group and irbesartan group after treatment,was statistically-significant lower than model group(P<0.05).But HDL-C level of TZQ high-dose group,TZQ middle-dose group and irbesartan group after treatment was no obvious statistically-significant compared with model group(P>0.05).④The expression level of CTGFmRNA and TGF-β1mRNA of TZQ high-dose group,TZQ middle-dose group and irbesartan group after treatment,were statistically-significant lower than model group(P>0.05).Compared The CTGFmRNA and TGF-β1mRNA level of TZQ high-dose group with irbesartan group after treatment,there was obvious statistically-significant(P<0.05).Conclusion:TangZhiQing Granules can significantly cut down blood sugar,blood lipid levels of the high sugar,high fat diet and low-dose Streptozotocin-induced model rats of type 2 diabetes and has the effect of prevention and cure diabetic cardiomyopathy.The mechanism may be related to improve the metabolism of glucose and lipid,supress the expression of TGF-β1mRNA and lower the CTGF-mRNA level.
