The Effect Of Guben Fangxiao Decoction On TNF-α And IL-8 Of Asthmatic Model At Remission Stage

Objective:To investigate the effect of Guben Fangxiao Decoction(GBFXD) on TNF-a and IL-8 of asthmatic mice at remission stage.Methods:The asthma model was reproduced in the BALB/c female mice challenged by repeated inhalation of ovalbumin(OVA) and repeated intranasal instillation of RSV. Then the mice were randomly divided into 8 groups to be treated with normal group, model group, prevention group, three doses(17.1g/kg,34.2g/kg and 68.4g/kg)of GBFXD group, montelukast sodium group, dexamethasone group(each group of 15) per day for 28 days. Samples for inspection at the end of the dosing after 24 hours. Observe the general activities of each mice group,observe the lung morphometry and histopathology by hematoxylin and eosin (HE) stain. Real-time quantitative polymerase chain reaction(RT-PCR) and ELISA assay were employed to confirm the expression of TNF-a and IL-8 in the lungs and BALF at mRNA and protein levels, respectively. Finally data analysis and processing by using statistical methods.Results:1. In normal group, alveolar structure in mice is clear, no significant alveolar inflammatory secretions appeared in alveolar cavities.The model group, the alveolar wall were damaged and a large number of inflammatory cell infiltration around. Compared with the model group, all of the administration groups significantly reduced airway inflammation, and inhibited mucosal edema and mucus secretion.2.The PCR results shown that:compared with the normal group, the levels of mRNA TNF-a, IL-8 of the model group was significantly higher. Compared with the model group, all of the administration groups could down regulate the level of TNF-a and IL-8 except the level of mRNA TNF-a of the low-dosed GBFXD group.3. The ELISA results shown that:the levels of protein expression changes in administration groups were lower than the model group,they all showed obvious difference. The low-dosed GBFXD group difference is not obvious.Conclusion:1. TNF-a and IL-8 were important inflammatory cytokines in the pathogenesis of asthma, may regarded as one of laboratory parameters in judge the disease changes; 2. Guben Fangxiao Decoction could down regulate the expression of TNF-a and IL-8 in asthmatic mice, reduce airway inflammatory cells infiltration and prevent the occurrence of asthma.
