The Effect Of Guben Fangxiao Decoction On ADAM33Gene And TGF-βof Asthmatic Model At Remission Stage

Objective:To study the effect of Guben Fangxiao Decoction on ADAM33gene and TGF-(3of asthmatic mice at remission stage; to explore the influence of serum containing Guben Fangxiao Decoction to MRC-5cell stimulated by IL-4.Methods:Animal research:Mice were sensitized by intraperitoneal injection with0.2mL0.05%OVA on dl,7,14except the controls. Then the mice were exposed to2.5%OVA in a atomizer for30minutes per day from d15to d21, followed by atomization every two days for3weeks. Then the models were divided to4groups to be treated with Guben Fangxiao Decoction (34.5g/kg), montelukast sodium (2.6mg/kg), astragalus polysaccharide (0.3g/kg) and saline (20ml/kg) per day for28days. The lung and BALF were needed to detect the expression of ADAM33and TGF-p.Cell research:Cells were synchronized without serum for8h in the logarithmic growth phase, followed by the stimulation of each concentration of IL-4for4h or24h; Cells were stimulated by the concentration of100ng/ml of IL-4for24h, followed by the addition of serum containing Guben Fangxiao Decoction, montelukast and saline. Cells were collected to be detected the level of ADAM33.Results:Animal research:The level of ADAM33and TGF-β of the model group was significantly higher than the control; All of the administration group could down regulate the level of ADAM33and TGF-β, although the level of ADAM33was still higher than the normal group, however, the level of TGF-β could be reduced to the normal.Cell research:The level of ADAM33was significantly reduced after the stimulation of each concentration of IL-4for4h, except for the group of lng/ml, while the level of ADAM33was significantly higher after the stimulation of IL-4for24h, except for the group of0. lng/ml and10ng/ml; However, the expression of ADAM33was down regulated in the group of Guben Fangxiao Decoction and montelukast after cells were stimulated by the concentration of100ng/ml of IL-4for24h.Conclusion:There was a specific relationship between ADAM33and asthma; Guben Fangxiao Decoction could down regulate the expression of ADMA33and TGF-β in asthmatic mice; IL-4act as a double role on ADAM33.
